摘 要:目的观察补充富含n-3多不饱和脂肪酸(PUFA)的亚麻籽油对高脂诱导肥胖小鼠血糖和胰升血糖素样肽-1(GLP-1)水平的影响。方法将30只雄性C57BL/6J小鼠分为正常饲料(ND)、高脂喂养(HFD)和亚麻籽油(FOD)组,每组10只,喂养8周后,行腹腔注射葡萄糖耐量实验(IPGTT)、胰岛素耐量实验(ITT)和GLP-1分泌实验。结果 FOD组FPG、胰岛素水平低于HFD组(P<0.01)。IPGTT结果显示,ND、FOD组120min血糖曲线下面积(AUC)低于HFD组(P<0.01)。ITT结果显示,HFD、FOD组120min胰岛素AUC比较差异无统计学意义。在等剂量亚麻籽油灌喂30min后,HFD、FOD组血浆GLP-1水平升高,且FOD组血浆GLP-1水平较HFD组高(P<0.01)。结论在高脂饮食的基础上长期补充亚麻籽油可改善小鼠血糖水平及IR,其机制可能是通过促进肠道分泌GLP-1实现的。Objective To explore the effects of flaxseed oil rich in n‐3 fatty acid on plasma glucose level and GLP‐1 secretion in C57BL/6J mice induced by high lipid dietary. Methods A total of 30 C57BL/6J mice were divided into normal diet (ND) group ,high fat diet (HFD) group ,and flaxseed oil diet (FOD ) group.After 8 weeks of treatment ,intra‐peritoneal glucose tolerance test (IPGTT ) ,insulin tolerance test (ITT) and GLP‐1 secretion were measured. Results Compared with HFD group ,FPG and insulin level were lower in FOD group (P〈0.01).The IPGTT showed that the AUC of glucose was lower in FOD group than in HFD group (P〈 0.01).The ITT demonstrated that the AUC of insulin was not significant different between FOD group and HFD group.At 30 min after intragastric administration of the same dose of flaxseed oil in three groups ,GLP‐1 secretion were increased in FOD and HFD group ,and FOD group was higher (P〈0.01). Conclusion Long term supplement of flaxseed oil in C57BL/6J mice induced by high lipid diet can improve the glucose level and IR.Further studies were needed to testify the possible underlying mechanism of stimulating the secretion of GLP‐1.
