摘  要:目的研究香菇多糖(lentinan,LNT)联合顺铂(DDP)对荷瘤小鼠实体瘤中S-100蛋白表达的影响。方法体外培养Lewis肺癌小鼠肿瘤细胞,接种健康BALB/c小鼠,荷瘤成功后予以治疗,实验分组为:荷瘤组,LNT组,DDP组以及LNT+DDP组。观察小鼠一般情况,并采用免疫组化法检测S-100蛋白表达,RTPCR检测S-100基因表达。结果肿瘤组织内S-100蛋白和m RNA表达水平LNT组、DDP组或LNT+DDP组比荷瘤组增多(<0.05),LNT+DDP组比LNT组和DDP组增多(<0.05)。结论 LNT同DDP联合使用上调S-100蛋白和基因的表达有协同作用。Objective To study the effects of lentinan(LNT) combined with cisplatin(DDP) on the expression of S-100 protein and m RNA in Lewis lung carcinoma-bearing mice. Methods Tumor cells of Lewis lung carcinoma were cultured in vitro and inoculated into healthy BALB/C mice. The mice were randomized into tumor-bearing group, LNT group, DDP group and LNT+DDP group. The expression of S-100 protein and m RNA was detected by immunohistochemical and RT-PCR respectively. Results The expression levels of S-100 protein and m RNA were significantly higher in LNT group, DDP group or LNT+DDP group than in tumor-bearing mice group(P〈 0.05), lower in LNT group or DDP group than in LNT+DDP group(P〈 0.05). Conclusion LNT and DDP have synergistic effect for the upregulated expression of S-100 gene and protein in Lewis lung carcinoma.
