摘  要:以小鼠为实验动物,探究1-脱氧野尻霉素(DNJ)在动物体内促进脂质代谢的作用机制以及作用途径的性别差异。分别按照0.002、0.004和0.008 mg/(g·d)的剂量给正常小鼠灌胃DNJ 40 d后,用ELISA试剂盒测定试验组不同性别小鼠肝脏中的乙酰辅酶A羧化酶(ACC)活性、肉碱棕榈酰转移酶(CPT-1)活性,血清中的脂联素(ADP)含量、内脂素(visfation)含量、游离脂肪酸(FFA)含量。3个DNJ灌胃剂量组雌、雄小鼠的部分脂质代谢指标出现变化:雌鼠的ACC活性分别降低了5.85%、7.79%、10.08%,ADP含量分别升高了0.04%、0.25%、7.27%,FFA含量分别下降了9%、12%、19%;雄鼠的CPT-1活性分别上升了4.2%、8.1%、21.9%,ADP含量分别升高0.83%、4.17%、23.39%,内脂素含量分别升高1.4%、7.6%、10.2%,FFA含量分别下降了6.97%、7.81%、12.47%。研究结果提示DNJ对不同性别小鼠的脂代谢影响途径不尽一致:通过降低雌性小鼠的ACC活性和FFA含量以及提升ADP含量来抑制脂肪酸合成;通过提升雄鼠的CPT-1活性和ADP含量,降低FFA含量来促进脂肪酸分解。In this paper,mice were used as animal model to explore the functional mechanism and effect of 1-deoxynojirimycin( DNJ) on lipid metabolism of femal and male mice. At 40 d after administration of DNJ at dosage of 0. 002,0. 004 and 0. 008 mg /( g·d),the activities of acetyl-Co A carboxylase( ACC) and carnitine palmitoyl transferase 1( CPT-1) in liver and the contents of adiponectin( ADP),visfation and free fatty acid( FFA) in serum of normalfemale and male mice were measured by ELISA kit. Some indexes of lipid metabolism in male and female mice were changed after oral administration of DNJ. In female mice of low,middle and high dosage group,the activity of ACC was decreased by 5. 85%,7. 79% and 10. 08% respectively; the content of ADP was increased by 0. 04%,0. 25% and 7. 27% respectively; the content of FFA was decreased by 9%,12% and 19% respectively. In male mice of low,middle and high dosage group,the activity of CPT-1 was increased by 4. 2%,8. 1% and 21. 9% respectively; the content of ADP was increased by0. 83%,4. 17% and 23. 39% respectively; the content of visfation was increased by 1. 4%,7. 6% and 10. 2% respectively; the content of FFA was decreased by 6. 97%,7. 81% and 12. 47% respectively. The above results indicated that the effect of DNJ on lipid metabolism of female and male mice were different. DNJ inhibits fatty acid synthesis in female mice by reducing ACC activity and FFA content,and by increasing ADP levels. On the contrary,DNJ promotes fatty acid decomposition in male mice by increasing CPT-1 activity and ADP content,and by reducing FFA content.
