高效液相色谱法测定酵母浸粉中维生素B_6的含量

摘  要:建立了高效液相色谱法测定酵母浸粉中维生素B6含量的方法。色谱条件为Symmetry-C18柱(3.9 mm×150mm,5μm);流动相:A相为甲醇,B相为含0.05 g/L的EDTA和1.1 g/L的庚烷磺酸钠的2.5%的乙酸溶液(p H 3.40),以V(A)∶V(B)为1∶6比例等度洗脱;流速为0.8 m L/min;柱温为30℃;检测波长为280 nm;样品分析时间为10 min。结果表明:维生素B6的线性范围为0.10~100.00μg/m L(r=0.999 9)。该方法检出限位16.0μg/100 g,定量限为50.0μg/100 g。日间和日内精密度均小于15%,低、中、高三种浓度下酵母浸粉样品中维生素B6的提取回收率分别平均为93.16%±2.00%、96.70%±2.08%、93.76%±1.79%(n=4)。此法操作简便、快速、准确,可作为酵母浸粉中维生素B6含量的检测方法。The purpose was to establish a method for determining the content of vitamin 136 in yeast extracting powder using HPLC. The detection conditions were: using Symmetry-C18 column (3.9 mm×150 mm, 5 μm), isocratic elution (V(A) : V(B)=1 : 6) and mobile phase A: methanol, phase B: 2.5% acetic acid solution containing 0.05 g/L EDTA and 1.l g/L sodium heptanesulfonate (pH 3.40), flow rate: 0.8 mL/min, detection wavelength: 280 nm, sample analysis time was 10 rain. The linear ranges of vitamin B6 were 0.10-100 μg/mL (r=0.999 9), The liunt of detection was 16.0 lag/100 g, and the liunt of quantification was 50.0 pg/100 g. relative standard deviation was less than 15%, recovery of the three concentration of vitamin B6 were 93.16%±2.00% , 96.70% ±2.08% and 93.76%±1.79% (n=4). The method was simple, rapid and reliable. It can be used in determination of vilamin 136 in yeast extracting powder.
