摘  要:为了研究酶解法提取米糠蛋白的工艺,试验采用Box-Behnken试验设计和响应面法,以脱脂米糠为原料,使用碱性蛋白酶和中性蛋白酶水解提取米糠蛋白,研究了不同水解时间、液料比、p H值、温度、加酶总量、加酶比例对米糠蛋白提取率的影响。结果表明:当液料比为4∶1、加酶总量为0.75%、碱性蛋白酶与中性蛋白酶用量之比为2∶1时,经碱性蛋白酶在p H值为10、温度为45℃的条件下水解3 h,再用中性蛋白酶在p H值为6.5、温度为45℃条件下水解1 h,米糠蛋白的提取率最高可达84.63%。说明由响应面法优化得到的工艺参数比较准确,具有实用性。To study the extraction process of rice bran protein by enzymatic hydrolysis, the alkali protease and neutral protease hydrolysis were used to extract rice bran protein using Box - Behnken experimental design and response surface methodology (RSM) with the defatted rice bran as material. The effects of different hydrolysis time, liquid -material ratio, temperature, pH value, the total amount of additive enzymes, and the ratio of additive enzymes on the extraction rates of rice bran protein were studied. The results showed that the alkaline protease was used to hydrolyze for 3 h at the pH value of 10 with the temperature at 45 ℃ when the liquid - material ratio was 4:1, the total amount of additive enzymes was 0. 75%, and the ratio of the dosages of alkali pretease and neutral pretease was 2:1 ; and then the neutral protease was used to hydro- lyze for lh at the pH value of 6.5 with the temperature at 45 ℃, the extraction rate of rice bran protein could reach 84.63%. The results indicate the extraction process parameters optimized by RSM are more accurate, and have practical significance.
