摘 要:目的检测miR-33、miR-211、miR-34a和miR-19a在氯化锂-匹罗卡品致痫大鼠海马区脑组织不同研究时点的表达水平,探讨其在癫痫发生发展过程中的作用。方法建立氯化锂-匹罗卡品癫痫大鼠模型,应用实时荧光定量PCR技术检测癫痫大鼠海马区脑组织不同研究时点miR-33、miR-211、miR-34a和miR-19a的表达量,并和对照组比较,对其差异进行分析。结果与对照组相比,miRNA-33在SE后1 h的表达水平明显降低(P<0.05),在SE后1 d和21 d表达水平明显升高(P<0.05),且SE后1 d表达量增高5.32倍;miRNA-211在SE后1 h的表达水平明显增高(P<0.05),且表达量增高3.56倍;在SE后1 d和60 d表达水平明显降低(P<0.05),在SE后60 d表达量降至0.17倍;miR-34a在SE后1 d和21 d的表达水平明显增高(P<0.05),在SE后21 d表达量增高3.12倍;miR-19a在SE后1 d的表达水平明显增高(P<0.05),且表达量增高4.32倍;在SE后60 d表达量明显下降(P<0.05),表达量降至0.39倍。结论 miR-33、miR-21、miR-34a、miR-19a在癫痫大鼠同一研究时点海马区脑组织中表达量存在差异,同一miRNA在不同研究时点海马区脑组织中的表达水平也有不同,提示各miRNA可能参与癫痫不同时期的调控。Objective To investigate the expression of miR-33、miR-211、miR-34a、miR-19 a in lithium-pilocarpine epileptic rat hippocampus and select the important time points in epileptogenesis. In order to explore the role of miRNAs in epileptic mechanisms. Methods Quantitative real-time PCR had beenwas used for detection of the expressionale status of miR-33、miR-211、miR-34 a and miR-19 a. By comparison of the expression status of those miRNAs at different time points in hippocampus of epileptic rat or normal rats hippocampus,it was possible to study the differential expression of miRNAs at different time points by statistic analysis. Results The results indicated that the expression of miR-33 had increased at 1day and 60 days post status epilepticus( SE) and declined at 1 hour after SE( P〈0. 05). MiR-211 had exhibited increased an increased expression level at 1 hour post SE,it had decreased at 1 day and 60 days post SE( P〈0. 05). MiR-34 a had exhibited an increased an expression level at 1 day and 60 days( P〈0. 05). MiR-19 a had exhibited an increased an expression level at 1 day post SE,it had decreased at 60 days post SE( P〈0. 05). Conclusion The expression change of miR-33、miR-211、miR-34a、miR-19 a in epileptogenesis at different time points had indicated their involvement in epileptic mechanisms which deserves to be studied and explored in future.
