摘  要:目的研究黄独乙素对人胃癌细胞SGC-7901增殖的影响及相关的作用机制。方法用RPMI-1640培养液对SGC-7901细胞进行传代培养,用不同浓度(0~160μg/m L)黄独乙素分别对SGC-7901细胞进行处理,应用CCK-8法检测细胞的增殖率;光学显微镜观察细胞形态学改变;Western blot检测细胞周期蛋白Cyclin D1表达的变化。结果黄独乙素以计量依赖方法抑制胃癌细胞生长,抑制率最高达57.26%;细胞形态发生明显改变;Cyclin D1蛋白水平表达显著下调。结论黄独乙素能显著抑制SGC-7901细胞的增殖,其机制可能与下调细胞周期蛋白Cyclin D1的表达有关。Objective To investigate effects of Disobulbin B (DB) on the proliferation of human gastric carcinoma cell line SGC-7901 and explore the possible mechanism. Methods SGC-7901 cells were treated with different concentration (0-160 pg/mL) of DB. Cell proliferation was detected by CCK-8 assay; cell morphology changes were observed under optical microscopy; The expression of CyclinD1 was detected by Western blot. Results DB inhibited the growth of gastric cancer cells in a does-dependent manner and the highest inhibition rate was 57.26%.The structure of SGC-7901 cells changed greatly. The protein expression level of cyclinD1 was reduced. The effects were increased with the increase of the DB concentration. Conclusion DB could inhibit cell proliferation in SGC-7901 cells by down-regulating the expression of CyclinD 1.
