摘  要:目的研究紫甘薯提取物(purple sweet potato extracts,PSPE)对更年期大鼠生理状态的影响。方法以11~12月龄雌性SD大鼠作为更年期模型,并以3~4月龄雌性大鼠作为中青年对照,对照组喂饲正常饲料,各剂量组大鼠连续饲喂不同剂量的PSPE,8w后观察大鼠的体重变化,计算子宫、卵巢系数,检测血清中雌二醇(E2)、促卵泡激素(FSH)、促黄体激素(LH)含量及子宫组织中雌激素受体(ER)水平。观察PSPE对卵巢组织形态的影响,并采用实时荧光定量PCR法测定子宫ER-α和ER-βm RNA相对表达量。结果与中青年对照组相比,更年期对照组大鼠呈现更年期的改变。而与更年期对照组相比,PSPE可以减缓更年期大鼠体重的持续上升,中剂量PSPE组大鼠的子宫、卵巢系数显著较高,高剂量PSPE组大鼠的血清FSH、LH水平明显较低,PSPE各剂量组大鼠的子宫组织中ER水平较高,卵巢生理学形态得到改善,子宫组织ER-α、ER-βm RNA相对表达量也明显较高。结论 PSPE可以调节大鼠内分泌功能,改善更年期的生理状态。Objective To investigate the effects of purple sweet potato extract(PSPE) on physiological status of the aging climacteric rats. Methods Female SD rats aged 11~12 months were selected as the climacteric model, whereas those aged 3~4 months were treated as the young control. Both the young and climacteric control rats were fed a normal diet and the climactic rats in three PSPE groups were fed the diets supplemented with low, medium and high dose PSPE, respectively. After 8 weeks of feeding, body weight, indices of uterus and ovary, levels of serum estradiol(E2), follicle stimulating hormone(FSH), luteinizing hormone(LH) and expression of estrogen reception(ER) in the uterus were measured. Histomorphologic structure of ovary was checked. The m RNA expressions of the ER-α and ER-β in the uterus were assessed by fluorescent real-time polymerase chain reaction. Results Compared to the young control rats, the rats in climactic control group showed typical climactic changes. Compared to the rats in the climacteric control group, the increase in body weight in climactic rats was slowed down after the treatment of PSPE. Additionally, the indices of uterus and ovary were increased in medium dose PSPE group, and the levels of serum FSH and LH were significantly decreased in high dose PSPE group. In three PSPE groups, the ER level was increased and the histomorphologic structure of uterus improved. Meanwhile, the m RNA expressions of ER-α and ER-β in uterus were significantly increased after PSPE treatment. Conclusion PSPE can improve the physiological status in aging climacteric rats by regulating the endocrine function.
