摘 要:为了提高漆斑菌Myrothecium sp.IMER1的胆红素氧化酶(bilirubin oxidase,BOX)产量,分析了其液体和固体发酵产酶的特性。首先分析了该菌株液体和固体发酵产BOX的时间变化曲线,对其所产的胞外酶进行了活性电泳,进一步分析了其液体产酶的模型和宏观动力学内容。结果表明,菌株IMER1在液体发酵中第5天的产BOX酶量最大,其固体发酵产酶量最大时间也是在第5天,但是,其固体发酵能持续高产BOX。从BOX的底物活性显色中可知,在固体和液体发酵的粗酶液中都能检测到BOX,并且没有同工酶出现;而BOX生物合成模式为中期合成型,通过数据计算和模拟得出了产酶动力学方程。In order to improve Myrothecium Myrothecium sp.IMER1 bilirubin oxidase(bilirubin oxidase, BOX)production, analysis of its liquid fermentation and enzymatic properties of solids. Firstly, the strain liquid and solid fermentation BOX time curve, were active electrophoresis their extracellular enzymes produced by further analysis of its liquid enzyme production model and macro dynamics content. The results showed that the strain in liquid fermentation IMER1 middle BOX enzyme 5 days maximum, its solid fermentation time is the maximum amount of enzyme in the first five days, however, it can continue to yield solid fermentation BOX. BOX substrate activity seen from the color, the crude enzyme solution in solid and liquid fermentation can be detected in the BOX, and no isozymes appear;and the mid-BOX biosynthesis mode synthetic, calculation and simulation data obtained by out of the production of the enzyme kinetics.
