摘 要:模拟正常人饮酒习惯,探讨摄入某品牌白酒后,不同时长及不同剂量对大鼠血糖、胰岛B细胞表达胰岛素(Ins)及血清Ins的影响。采用正常成年雄性SD大鼠,随机分为正常对照组及实验组。其中实验组分为4周组(4周)、8周组(8周)、12周组(12周)3个时间点;每个时间点又分为低剂量组(L组,0.8 m L/kg·d)、中剂量组(M组,1.6m L/kg·d)、高剂量组(H组,2.4 m L/kg·d)。实验组大鼠每日11AM及5PM各灌胃1次,正常对照组不予处理。分别于第4周末、第8周末、第12周末测空腹血糖,采集血清,取胰尾组织。采用放射免疫测定法、免疫组化SABC法、图像分析及形态计量法进行研究。结果表明,各实验组大鼠空腹血糖水平及大鼠胰岛面数密度与正常对照组比较,差异无统计学意义(P>0.05);与正常对照组比较,除8周(H)组和12周(L)组稍有升高外,其余实验各组血清Ins、胰岛B细胞的免疫染色强度及平均光密度的差异无统计学意义(P>0.05)。在本实验设定的剂量和时程内用该白酒灌胃,大鼠血糖未见明显异常;对胰岛B细胞表达和面数密度、大鼠血清Ins水平无明显影响。In this study, simulating liquor drinking habits of normal people, we explored the expression changes of insulin in pancreatic islet B cells and serum insulin levels in rats after consumption Baijiu(liquor) of different time period and different dose. Normal male adult SD rats were randomly divided into control group and experiment groups. Experiment groups included 4 weeks group(4w), 8 weeks group(8w) and 12 weeks group(12w);each group were further divided into low dose group (L, 0.8 mL/kg·d), middle dose group (M, 1.6 mL/kg·d) and high dose group (H, 2.4 mL/kg·d). Rats in experiment groups were given by gavage twice a day(11 AM and 5 PM) with Baijiu(liquor), while rats in con-trol group received no special treatment. At the end of the 4th, 8th and 12th week, the rats’fasting blood glucose was measured, serum samples and pancreatic tail tissue were collected. Samples were analyzed by radioimmunoassay, immunohistochemical SABC, image analysis and mor-phometry methods. The results showed that, there was no statistically significant difference in fasting blood glucose level and pancreatic islet numerical density on area among experiment groups and control group (P〉0.05). Compared with control group, the immune staining intensity of pancreatic islet B cells and serum insulin levels of 8w(H) group and 12w (L) group increased slightly(P〈0.05). The treatment of gavage with 54%vol Baijiu(liquor) at the dosage and time period in this experiment had no significant effect on fasting blood glucose, immune staining in-tensity and numerical density on area of pancreatic islet B cells, and serum insulin levels of rats.
