摘 要:目的:应用体视学方法定量研究跑步对早期APP/PS1转基因阿尔茨海默病(AD)小鼠海马CA1和齿状回区域内神经元的影响.方法:将6月龄雄性APP/PS1转基因AD小鼠,随机分为跑步组和未跑步对照组.用Morris水迷宫检测2组小鼠的空间学习记忆能力,然后用体视学方法定量研究海马CA1和齿状回区域的体积和神经元的数量.结果:与对照组相比,跑步组AD小鼠Morris水迷宫的逃避潜伏期明显缩短,平台所在象限时间百分比和穿越平台次数均显著增加.体视学研究显示,跑步组AD小鼠海马CA1和齿状回的体积分别比对照组显著增大42.8%和27.3%,并且跑步组AD小鼠海马CA1和齿状回区域内的神经元数量分别比对照组显著增加61.8%和62.2%.结论:跑步能延缓早期AD小鼠空间学习记忆能力的下降;并且可降低AD小鼠海马CA1和齿状回区域内神经元的死亡.Objective: To investigate effects of the exercise on the number of neurons in the hippocampus CA1 subfield and in the dentate gyrus (DG) of early transgenic Alzheimer's disease (AD) mice with the stereological method. Methods: Twenty 6-month-old male APP/PS1 transgenic mice were randomly divided into a running group and a control group (10 mice per group). The running group mice ran for four months, and the control group mice were housed in standard condition without running. Then, the behaviors of the mice from both groups were assessed with the Morris water maze test, and the volumes of the hippocampal CA1 subfield and DG and the number of the neurons in the CA1 and DG were investigated with the stereological methods. Results: Compared to the control group, the escape latency in the Morris water maze test mice of the running was significantly shortened, and the time percentage of platform quadrant and the times of crossing the platform were significantly increased. Volume of CA1 subfield and DG was significantly increased by 42.8% and 27.3%, and the number of neurons in CA1 subfield and DG was significantly increased by 61.8% and 62. 2% seperately in the running mice. Conclusion: The present results suggests that exercise can delay the decline in the learning and spatial memory ability and the neuron death in the CA1 and DG in early AD mice.
