摘 要:目的探讨黄曲霉毒素B1(AFB1)对体外正常人胚肝细胞(L-02细胞)的急性损伤作用。方法 L-02细胞体外培养,随机分为9组,空白对照组(不加其他试剂),溶剂对照组(加入二甲基亚砜),不同浓度AFB1染毒组(分别加入5、10、20、40、80、160、320 mg/L)。细胞培养24 h后观察各组细胞的形态学变化,并应用MTT法检测各组细胞吸光度值(A),计算细胞毒性指数(CI)。结果培养24 h后,镜下观察发现随着AFB1染毒剂量增大,L-02细胞皱缩变形和凋亡逐渐增多,当浓度达到80 mg/L时可见大量漂浮细胞和细胞碎片。各组细胞吸光度值比较差异有统计学意义(P<0.05),80、160、320 mg/L AFB1染毒组的吸光度值均低于空白对照、溶剂对照组(P均<0.05),并且随着AFB1染毒剂量提高,细胞毒性指数逐渐增高。结论 AFB1染毒浓度大于80 mg/L时,对L-02细胞有显著毒性作用。Objective To explore the acute injury of human embryo hepatocytes (L-02 cell) in vitro induced by aflatoxin B1 (AFB1 ). Methods L-02 cells were cultured in vitro and randomly divided into 9 groups,blank control group (without other reagents) , solvent control group( with dimethyl sulfoxide ) ,AFB1 exposed groups(5,10,20,40,80,160,320 mg/L). The morphological changes were observed in all groups 24 hours after the cell culture,methyl thiazol tetrazolium(MTT) assay was used to detect the cell absorbance value (A) of the cells in each group,and the cytotoxicity index(CI) was calculated. Results After 24 hours of culture, the microscopic observation revealed the deformation and apoptosis of L-02 cell gradually increased with increasing exposure dose of AFB1 ,a large number of floating cells and cell debris were observed when the exposure dose of AFB1 was 80 mg/L. There was significant difference in the cell absorbance value among all groups(P 〈0. 05) ,the absorbance value was lower in the AFBl exposed groups of 80,160,320 mg/L compared with blank control and solvent control groups(all P 〈 0. 05), and the cytotoxicity index increased gradually with increasing exposure dose of AFB1. Conclusion AFB1 has significant toxic effects on L-O2 cells when its exposure dose is higher than 80 mg/L.
