摘 要:目的:利用Design-expert 8.0响应面分析法RSM(Response Surface Methodology)对小花鬼针草黄酮类化合物提取工艺进行优化。方法:在提取温度、提取时间,料液比,提取浓度4个单因素实验基础上选取实验因素与水平,根据中心组合(Box-Benhnken)实验设计原理采用4因素3水平的响应面分析法,以获得多元二次线性回归方程,以黄酮提取率为响应值作响应面和等高线。结果:提取时间和液料比2个因素以及二者交互作用对小花鬼针草的总黄酮物质得率影响差异显著,确定了超声波浸提小花鬼针草最佳工艺条件:温度=61.83℃,时间=2.40h,液料比=15.67,乙醇浓度=75%,决定系数R2=0.72520表明方程拟合度高,二次回归方程模型差异显著(P=0.04),失拟项不显著(P=0.11)。结论:小花鬼针草黄酮类化合物的提取率可达5.289,基于响应面法分析所得的优化提取工艺参数准确可靠具有实用价值。In this work, ultrasound technology was used for the extraction of total fl avonoids from Bidens parvifl ora Willd. Response surface methodology(RSM), based on a Box-Benhnken design(BBD), was used to optimize the effects of processing parameters on total fl avonoids yields. The parameters were extraction temperature(A), extraction time(B), ratio of liquid to solid(C) and ethanol concentration(D). The statistical analysis indicated that the parameters of extraction time and ratio of liquid to solid, and the interaction effects among them had signifi cant effects on the yield of fl avonoids from Bidens parvifl ora Willd. The optimum extraction conditions were determined as follows: extraction temperature 61.83 ℃ extraction time 2.40 h, ratio of liquid to solid 15.67 m L/g, ethanol concentration 75%. Model coeffi cient of determination R2=0.72520 indicates that the regression equations fit high, quadratic regression model differs significantly(P=0.04), and no significant lack of fit(P=0.11). Under these conditions, the experimental yield agreed closely with the predicted yield(5.289%).
