补肾壮阳类中成药中非法添加物PDE_(5)抑制剂的分离鉴定

摘　　要：目的研究补肾壮阳类药物中非法添加物的提纯与结构鉴定,为国内研究补肾壮阳中成药及保健品中非法添加物提供技术支持和相关谱图数据。方法使用薄层色谱、柱色谱、高效液相色谱对非法添加物进行分离和提纯;使用核磁共振、质谱确定非法添加物的化学结构。结果提纯得到黄色针状结晶,纯度为97%,化学结构确定为5-{5-[4-(2-羟乙基)哌嗪磺酰]-2-正丙氧基苯基}-1-甲基-3-正丙基-1H-吡啶并[4,3-d]嘧啶-7(6H)-硫酮。结论该化合物鲜有报道,需密切关注。AIM To purify and identify the adulterated PDE_(5) in healthy products of promoting sexual enhance-ment,and to provide technical support and related spectral data for the supervision and inspection.METHODS The separation and purification of the adulterated was performed using TLC(thin layer chromatography),CC(col-umn chromatography)and HPLC(high performance liquid chromatography).NMR(nuclear magnetic resonance)and MS(mass spectrometer)were used for the determination of the chemical structure.RESULTS Yellow nee-dle crystal,whose purity was up to 97%,was obtained,and its structure was proved to be 5-{5-[4-(2-hydroxyethyl)piperazin)sulfonyl]-2-propoxyphenyl/-1-methyl-3-propyl-lH-pyrazolo[4,3-d]pyrimidine-7(6H)-thione.CONCLUSION The constituents obtained from Buhe Capsules are rarely reported,so we should pay close attention to it.
