摘 要:[目的]观察N-甲基-N′-硝基-N-亚硝基胍(MNNG)不同给药剂量及给药途径对大鼠胃黏膜组织病理学的影响。[方法]将120只雄性SD大鼠随机分为3组,空白组40只,模型Ⅰ组40只、模型Ⅱ组40只。模型Ⅰ组采用100μg/ml的MNNG自由饮用,联合雷尼替丁饲料、饥饱失常、烫热高盐饮食造模;模型Ⅱ组在模型Ⅰ组造模方法的基础上,每日以0.017mol/L的MNNG溶液1ml/只灌胃,连续28周。比较各组大鼠一般情况、死亡率、组织病理学变化及PLGC诱变率。[结果]模型Ⅰ组大鼠死亡率(5%)低于模型Ⅱ组(23%);模型Ⅰ组PLGC诱变率(84.6%)明显高于模型Ⅱ组(55.6%),差异有统计学意义(P<0.05);模型Ⅱ组鳞状上皮异型增生、鳞癌发生率分别16.7%、50%,模型Ⅰ组和空白组均未见前胃黏膜组织学变化。[结论]MNNG自由饮用联合灌胃、雷尼替丁饲料、饥饱失常、烫热高盐饮食能成功诱导大鼠胃癌前病变模型;但联合大剂量MNNG灌胃使PLGC诱变率降低,死亡率上升,并出现明显前胃鳞状上皮病变。[Objective]To observe the effect of different routes and doses of N-methyl-N-nitro N-ni- trosoguanidine(MNNG) administration on pathological changes of gastric mucosa in rats. [Methods] 120 male SD rats were randomly divided into three groups, control group (n = 40), model group Ⅰ (n = 40) and model group Ⅱ (n=40). The model group Ⅰwere given 100 μg/ml MNNG free drink and 0.03% ranitidine ,irregular diet and 56℃ 15 G sodium chloride for 28 weeks. The model group Ⅱ were given the above modeling plus gastric perfusion of 0. 017 mol/L MNNG. The general status, the mortality,pathological changes of gastric mucosa and mutagenic rate were compared in the three groups. [Results]The mortality rate in group Ⅱ (5%) was significantly lower than that in group Ⅱ(23%), while the mutagenic rate was observed higher in group I (84.6%) than in group Ⅱ (55.6%) in model group Ⅱ ;The incidence rate of hyperplasia and squamous carcinoma of forestomach in model group Ⅱ was 16.7% and 50%. The model group I and control group showed no abnormal changes in forestomach mucosa. [Conclusion]Free drink of MNNG combined with ranitidine, irregular diet, hot and high-salt diet induces precancerous lesions of gastric cancer (PLGC). High doses MNNG intragastric administration lowers the mutagenic rate, while increases the mortality rate,and the squamous epithelium changes of forestomach was induced.
