摘  要:目的 了解2010—2013年广州市食源性疾病监测副溶血性弧菌分离株的血清型别、抗菌药物耐药性、毒力基因携带情况以及分子分型特征。方法 对广州市2010—2013年食源性疾病监测中分离到的108株副溶血性弧菌进行血清分型、药敏试验以及耐热直接溶血毒素基因(tdh)、耐热相关溶血毒素基因(trh)的PCR检测,并进行脉冲场凝胶电泳(PFGE)分型。结果 2010—2013年广州市副溶血性弧菌的主要血清型为O3:K6(51株,47.2%)、O1:KUT(32株,29.6%)。药敏检测结果显示,分离株对氨苄西林(90.7%)和头孢吡肟(28.8%)耐药率较高,而对复方新诺明、氯霉素则100.0%敏感。多重耐药分析显示,2010—2013年分别有53.6%(15/28)、20.6%(7/34)、18.8%(3/16)、33.3%(10/30)的分离株同时耐受≥3种抗菌药物。毒力基因PCR检测显示,95.4%(103/108)的分离株为tdh+trh-,4.6%(5/108)的分离株为tdh-trh+。PFGE显示,108株副溶血性弧菌经NotⅠ酶切后的PFGE图谱可分为3个聚类,55个PFGE型别,相似值为60.2%~100%。优势血清型O3:K6和O1:KUT主要集中于聚类Ⅱ,O4:K8血清型主要集中于聚类Ⅲ。结论 2010—2013年广州市食源性疾病监测副溶血性弧菌优势血清型为O3:K6和O1:KUT型,菌株对多数抗菌药物仍然比较敏感,但存在多重耐药现象,多数菌株携带tdh基因,PFGE结果提示广州市食源性监测副溶血性弧菌存在遗传多样性。Objective To study the serotypes, antimierobial resistance, virulence genes and molecular characteristics of Vibrio parahaemolyticus isolated from foodborne diseases during 2010-2013 in Guangzhou. Methods A total of 108 Vibrio parahaemolyticus strains isolated from foodborne diseases were collected to serotype and antimicrobial resistance tests. PCR was used for the detection of tdh (thermostable direct hemolysin gene) and trh ( tdhrelated hemolysin gene). All the samples were analyzed by pulsed -field gel electrophoresis (PFGE). Results A total of 108 strains of Vibrio parahaemolyticus obtained from diarrhea patients with foodborne diseases were collected, among which O3 : K6 (51, 47.2% ) and O1 :KUT (32, 29.6% ) were the dominant serotypes. Vibrio parahaemolyticus isolated showed high resistance to ampicillin (90.7%) and cefepime (28.8%), but were 100.0% sensitive to cotrimoxazole and chloramphenicol. Multi - drug resistance was revealed in 53.6% (15/28) , 20.6% (7/34), 18.8% (3/16) and 33.3% (10/30) of isolated strains in 2010, 2011, 2012 and 2013, respectively. Results from virulence gene detection suggested that 95.4% (103/108) isolates belonged to tdh- trh+ , while 4.6% (5/108) isolates belonged to tdh- trh +. Results from PFGE a- nalysis suggested that 108 isolates were discriminated into 3 clusters and 55 different PFGE patterns with the similarity val- ue between 60.2% and 100.0%. The dominant serotype O3 :K6 and O1 :KUT seemed to gather in the cluster Ⅱ, while serotypc 04 : K8 in cluster Ⅲ. Conclusion 03 : 1(6 and O1 : KUT are the dominant serotypes in Vibrio parahaemolyticus strains isolated from foodborne diseases during 2010-2013 in Guangzhou. These strains show high sensitivity to most antibiotics, but with multi - drug resistance. Positive rate of tdh gene is high. PFGE analysis reveals genetic diversity is within the Vibrio parahaemolyticus strains of foodborne diseases in Guangzhou.
