摘  要:目的:优选牡丹皮炭的炮制工艺。方法:采用砂烫法,以5-羟甲基糠醛、没食子酸、儿茶素、丹皮酚含量的综合评分为指标,在单因素试验基础上,选择炮制时间、炮制温度、投料比为自变量,采用Box-Behnken响应面法优化牡丹皮炭的炮制工艺。结果:牡丹皮炭最佳炮制工艺为炮制时间9.0 min,炮制温度180.0℃,投料比25.5倍。5-羟甲基糠醛、没食子酸、儿茶素、丹皮酚质量分数分别为0.022 1%,0.023 3%,0.175 7%,0.042 0%,综合评分预测值97.67分,实测值95.01分。结论:优选的炮制工艺简便可行,预测性良好,充分体现了牡丹皮炭凉血止血、止血而不留瘀的作用特点。Objective: To optimize processing technology of Moutan Cortex carbon. Method: With the content of gallic acid, 5-hydroxymethyl furfural, catechin, paeonol as comprehensive evaluation index, based on single factor tests, herb-to-sand ratio, processing time and temperature were selected as independent variables, sand-scorch method was adopted, processing technology of Moutan Cortex carbon was optimized by Box-Behnken response surface methodology. Result: Optimum processing conditions were as follows: processing time of 9.0 min, processing temperature at 180.0 ~C , herb-to-sand ratio of 25.5: 1. The content of 5-hydroxymethyl furfural, gallic acid, catechin and paeonol were 0. 022 1% , 0. 023 3% , 0. 175 7% and 0. 042 0% , respectively. The measured value of composite score was 95.01, which was close to the predicted value of 97.67. Conclusion: This optimized processing technology of Moutan Cortex carbon is simple and feasible with good predictability, which fully embodies function characteristics of this herb.
