摘 要:为制备纯度大于90%以上的蛋黄磷脂酰胆碱产品,采用三氧化二铝柱层析法,以蛋黄粗磷脂为原料,食用级乙醇为洗脱剂,制备磷脂酰胆碱产品。通过单因素试验,研究了洗脱剂极性、洗脱剂流速、样品加样量对精制效果的影响。研究结果表明在洗脱溶剂为80%的乙醇溶液、洗脱液流速为0.3 mL/min、上样量为柱体积的3%条件下,得到磷脂酰胆碱含量为96.46%、回收率为95.78%的蛋黄磷脂酰胆碱产品。经研究证明此工艺是制备高纯度蛋黄磷脂酰胆碱产品较为有效的手段。In order to prepare high-purity egg yolk phosphatidylcholine, the purity of which is more than 90%, alumina column chromatography method was used, egg yolk crude phospholipids were used as raw materials and consumption alcohol was used as elution resolver to refine phosphatidylcholine products. Single-factor test was used to study the solvent concentration. The effects of the elution resolver flow and the rate of sample in the column volumes on the refining was studied. The result showed that the optimum purity condition was as following: the flow rate at 0.3 mL/min, solvent concentration 80% and the rate of sample in the column volume 3~/0. Though the technology, the egg PC products could be obtained in which the content of PC was 96.46% and the recovery rate was 95.78%. The process has more effective means on the preparation of high-purity egg yolk phosphatidylcholine products.
