摘  要:采用AB-8型大孔树脂、溶剂处理及重结晶相结合的方法对葛根异黄酮进行分离与部分纯化的工艺优化,并用紫外分光光度法测定异黄酮含量。结果表明,大孔树脂的最佳控制参数为:样液质量浓度75 mg/mL时上样量261 mL,样液pH 5~6,吸附流速20 mL/min,依次用4倍及3倍床体积的水、70%乙醇洗脱,流速为20 mL/min。结合溶剂处理及重结晶,所得产品的异黄酮含量可达5.448 1 mg/g,远高于传统的水提和醇提法。The process optimization of the extraction and partial purification ofPueraria lobata tuber isoflavones was conducted by using a method with a combination of AB-8-macroporous resin column chromatograpgy, solvent treatment and recrystallization, and the contents of isoflavones were assayed by a UV-Vis spectrophotometry. The results showed that the optimal control parameters for macroporous resin were: the solution conc of 75 mg/mL, the volume of 261 mL; pH 5--6, the flow rate of 20 mL/min. Elute with 4 and 3-folds of the resin bed volumes of water and 70% ethanol respectively, a flow rate of 20 mL/min. To continue with solvent treatment and recrystallization, the contents of isoflavone can be up to 5.448 1 mg/g, much higher than that with traditional methods by using water and alcohol respectively. The method is simple, and easy to promote.
