天然油脂和表面活性剂对发酵性丝孢酵母产油脂及脂肪酶的影响

摘  要:通过摇瓶发酵,研究了天然油脂(大豆油、猪油)和表面活性剂(Tween80、洗洁精)对发酵性丝孢酵母(Trichosporon fermentans)发酵生产油脂和胞内脂肪酶的影响。结果表明,大豆油对细胞产油和产酶的促进效果好于猪油,最适添加量为体积分数1.0%,发酵60h最大脂肪酶活力为347.5U/mL,发酵72h最大细胞油脂质量分数可达30.7%。低浓度表面活性剂对促进细胞产酶有利;Tween 80对细胞产油和产酶的提高效果好于洗洁精,最适添加量为体积分数0.5%,发酵60h最大脂肪酶活力为297U/mL,发酵72h最大细胞油脂质量分数可达30%。对于发酵性丝孢酵母,细胞内脂肪酶活力增加对油脂积累有促进作用。The effects of natural oils (soybean oil, lard) and surfactants (Tween 80, detergent) on the production of microbial lipid and intracellular lipase were studied in submerged fermentation by Trichosporon fermentan. The results showed that soybean oil is better for lipid and lipase production than lard. Lipase activity was up to 347.5 U/mL after 60 h fermentation and lipid content reached to 30.7% after 72 h fermentation by adding 1.0~ (volume fraction) soybean oil. Lipase production was improved in the presence of low concentration of surfactant. The production of Lipid and lipase was higher adding Tween 80 than detergent. When 0.5% (volume fraction) Tween 80 was added into fermentation medium, lipase activity was up to 297 U/mL after 60 h fermentation and lipid content reached 30% after 72 h fermentation. For Trichosporon fermentan, the improvement of intracellular lipase activity had a positive effect on microbial lipid accumulation.
