摘 要:目的女性受孕期间子宫内膜能量代谢正常与否,直接决定受精卵的种植成功率及胚胎正常发育。本研究拟探讨子宫内膜细胞线粒体DNA(mtDNA)缺失及能量变化与不孕症发生的关系。方法收集原发性不孕症(n=30)、继发性不孕症(n=27)及对照组(n=26)的子宫内膜组织,采用巢式多聚酶链反应(PCR)检测各组子宫内膜细胞中mtDNA缺失差异;采用实时荧光定量PCR(Real-time PCR)检测各组mtDNA拷贝数变化;最后,比较各组子宫内膜组织的ATP含量差异。结果巢式PCR检测显示,原发和继发性不孕症患者子宫内膜细胞mtDNA 4 977bp缺失率分别为86.67%和78.78%,均显著高于对照组(38.46%)(P<0.05);原发和继发性不孕症子宫内膜细胞mtDNA拷贝数明显增加,分别比对照组高48.49%和51.01%(P<0.01),而ATP含量显著下降,分别比对照组低57.00%和61.29%(P<0.01)。结论子宫内膜mtDNA缺失率增加,mtDNA拷贝数增加,引起子宫内膜能量代谢失调进而参与不孕症的发生。Objective: To investigated the relationship between endometrial mitochondrial DNA (mtDNA) deletions and female infertility. Methods: Endometrial tissues were collected from 30 primary infertility, 27 secondary infertility and 26 normal fertile women. The mtDNA deletions were detected by nested polymerase chain reaction(PCR). The mtDNA copy number was determined by real-time quantitative PCR, while ATP contents were detected by luciferin-luciferase assays. Results: The deletion rates of mtDNA 4 977 bp in primary(86.35%)and secondary infertile groups (78.07%) were significantly higher than that in normal fertile women (41.27%)(P〈0.05). The significantly higher mtDNA copy number and lower ATP contents were found in primary and secondary infertile women compared to normal fertile women(P〈0.01). Conclusion: The increased mtDNA deletion rate and high mtDNA copy number could lead to disorder of endometrial energy metabolism and female infertility.
