摘 要:为了缩短高山被孢霉(Mortierella alpina)产花生四烯酸(ARA)油脂发酵周期,以提高ARA油脂生产强度,主要研究了不同底物流加方式对M.alpina产ARA油脂的影响。考察分批发酵、残糖反馈补料分批发酵对ARA油脂发酵的影响,并进一步在残糖反馈补料发酵的基础上建立了一种反复补料分批发酵工艺。结果表明:与分批发酵相比,虽然细胞干质量和油脂浓度变化不显著,但是残糖反馈补料方式ARA生产强度从0.93提高至1.33g/(L·d)。在残糖反馈补料基础上反复补料分批发酵一共进行了4批,细胞干质量稳定在30 g/L左右,油脂含量稳定在50%左右,ARA含量分别达到42.81%、43.17%、42.30%和39.71%。To shorten the production cycle of arachidonic acid( ARA)-rich oil and increase the productivity,we studied the effects of different substrate feeding strategies on the production of( ARA)-rich oil by Mortierella alpina. Batch fermentation and residual glucose feedback fed-batch fermentation were studied,as well as a repeated fed-batch fermentation that was developed on the basis of residual glucose feedback fed-batch. Compared with batch fermentation,ARA productivity in residual glucose feedback fed-batch fermentation increased from 0. 93 to 1. 33 g /( L·d). Four batches were carried out for repeated fed-batch culture,in which the biomass remained stable around 30 g / L and lipid contents remained stable around 50%. ARA contents in total fatty acid were 42. 81%,43. 17%,42. 30%,and39. 71%,respectively.
