摘  要:以无甲醇诱导型基因重组人血清白蛋白的毕赤氏酵母(Pichia pastoris)NCY-1为实验菌株,在10 L机械搅拌通风发酵罐水平和3 L摇瓶水平的基础上研究了该菌株发酵过程中参数的变化规律及铵根离子(NH4+)对发酵液中重组人血清白蛋白(r HSA)的影响。研究结果表明:发酵液中的r HSA的含量随菌体浓度的增加而增加;30 h后菌体进入稳定期,此时流加10 g/L的葡糖糖有利于发酵液中r HSA的表达,流加后发酵液中的r HSA最高产量可达到185 mg/L;发酵液中存在活性较高的蛋白酶,在发酵后期分解r HSA,但发酵液中铵离子(NH4+)质量浓度为3.0 g/L时能较好的抑制r HSA的分解。Using non methanol induced recombinant Pichia pastoris NCY-1 which produce recombinant human serum albumin (rHSA) as experimental strain, the changing regulations of fermentation parameters and the effect of NH4^+ on the rHSA in fermentation broth were studied on the level of 10 L mechanically stirred tank and 3 L shake flask. According to the results, the output ofrHSA in fermentation broth increased with the increase of cell concentration. After 30 h, NCY-1 entered into stationary phase. At this time, the feeding of 10 g/L glucose could improve the expression of rHSA, and the rHSA production reached up to 185 mg/L. There was protease with high activity in the fermentation broth which can decompose rHSA in the late fermentation. The NH4^+ concentration of 3.0 g/L can inhibit rHSA decomposition effectively.
