摘  要:环境内分泌干扰物可对生物体的内分泌系统、生殖系统、免疫系统等的功能造成障碍,对机体产生可逆或不可逆的生物学效应。由于免疫系统的反应较机体其他系统更为敏感,因此免疫毒理学目前在食品安全和环境安全领域已成为重要的研究课题。本研究分别用50、100和200 mg/kg的双酚A(BPA)以及10 mg/kg雌二醇(E2)腹腔注射于体重约800 g的健康半滑舌鳎(Cynoglossus semilaevis),暴露14 d后,分别取各处理组的胸腺、肝脏、肾脏、脾脏和心脏等5种组织,通过实时荧光定量技术分析了BPA和E2对半滑舌鳎免疫系统关键转录调节因子(Ikaros)的影响。研究发现,50 mg/kg的BPA对Ikaros的表达量影响稍弱,只在胸腺和肝脏中出现了较为明显的表达水平的上调。而在100和200 mg/kg的BPA处理组中,Ikaros基因在胸腺、肝脏、肾脏和脾脏4种组织中的表达水平均出现了明显的增长。在10 mg/kg E2处理组中,Ikaros在4种组织中的表达亦均显著上调,相比较于对照组心脏中的表达水平,均上调20倍以上。结果说明,在高剂量的BPA或E2的胁迫下,Ikaros对内分泌干扰物的胁迫反应较为强烈,且其表达水平随BPA剂量的增加出现了明显的上调,表明其在内分泌系统中亦应具有特定的调控功能,但具体的调控机制尚需要进一步的研究。本研究可为鉴定Ikaros在半滑舌鳎内分泌系统中的调控功能提供参考数据。Environmental endocrine disruptors can cause disorders of the function of the endocrine system, reproduc- tive system and immune system, and produce adverse biological effects on the organisms. Since the reaction of im- mune system of the organisms are more sensitive than other systems, and therefore immune toxicology has become an important research topic in the field of food safety and environmental safety currently. By real -time quantitative PCR technique, the expression patterns of Ikaros gene, as a key transcription factor for the immune system, were analyzed in different tissues including thymus, liver, kidney, spleen and heart from C. semilaevis weighing about 800 g which was treated with 50 , 100 and 200 mg/kg Bisphenol A (BPA) or 10 mg/kg Estradiol (E2) by intraperitoneal injec- tion after 14 d. The results showed except 50 mg/kg BPA for only an obvious increase of gene levels in thymus and liver, Ikaros had a strong response to the stress of 100 and 200 mg/kg BPA, whose expression levels increased signifi- cantly in thymus, liver, kidney, and spleen tissues. In 10 mg/kg E2 treated group, the gene levels of Ikaros were up- regulated more than 20 times in four different tissues. This suggested that Ikaros was strongly responded under the stress of EDCs and dependent on concentrations of BPA. Moreover, it also suggested that Ikaros may have a particular function in the endocrine system. But the specific regulatory mechanism stilX needs further research. This study provides data to identify Ikaros function.
