消溃平液抗胃癌癌前病变的实验研究

摘　　要：[目的]通过动物实验观察消溃平液抗胃黏膜细胞突变、癌变的作用。[方法]1应用化学致癌快速筛选短期实验的细胞非程序合成(UDS)的方法,做大鼠体内和体外突变实验,观察消溃平液对化学致癌物MNNG诱导的大鼠外周血白细胞突变的阻断突变作用。2用小鼠90只,分为3个组,1组饮自来水,2组自由饮含150ug/ml致癌物—MNNG的自来水,3组自由饮含150ug/ml MNNG加含25mg/ml消溃平液自来水,2周后处死动物,胃切片病理诊断癌变。结果1体内诱导实验,大鼠经2周饲养诱导UDS值从4120.31±188.22降到3151.28±82.91,P<0.01;体外诱导血中白细胞UDS值由4187.22±592.42下降到3102.23±306.58,P<0.01;2小鼠饮含致癌物MNNG组30只中17只发生了胃黏膜癌变,而饮用MNNG同时加饮25ml/l消溃平液30只中5只发生癌变,抑癌率70.58%。[结论]消溃平液能抑制致癌物MNNG对大鼠外周血白细胞诱发突变的作用;对小鼠胃黏膜增生癌变有高效的抑制作用。[Objective]To observe the role of Xiaokuiping liquid in anti-mutation and anti-canceration by animal experiment.[Methods]（1）The effect of Xiaokuiping liquid on rat peripheral blood leukocytes,which was induced mutation by chemical carcinogens MNNG,was observed in rat mutation experiments in vivo and in vitro based on the method of unscheduled DNA synthesis（UDS）test of chemical carcinogenesis rapid screening short-term experiment.（2）90mice were divided into 3groups.No.1group mice were given tap water.No.2group mice were given 150ug/ml carcinogens-MNNG diluted with tap water.No.3group mice were given MNNG and 25 mg/ml Xiaokuiping liquid diluted with tap water.After 2 weeks,the animals were sacrificed and the stomach histopathology was examined to diagnose canceration.[Results]（1）In vivo induction experiments,the UDS value was decreased from 4120.31±188.22 to 3151.28±82.91（P〈0.01）.In vitro,the UDS value of blood leukocytes was decreased from 4187.22±592.42 to 3102.23±306.58（P〈0.01）.（2）17mice of No.2group were found canceration in gastric mucosae in contrast to the 5mice of No.3group.The tumor-inhibition rate was 70.58%.[Conclusion]Xiaokuiping liquid can inhibit the mutation of rat peripheral blood leukocytes induced by chemical carcinogens MNNG,and can effectively inhibit the hyperplasia and canceration of the mouse gastric mucosa.
