摘 要:为认识原核Mre B蛋白和真核actin蛋白结构相关性和进化关系,应用SWISS-MEDOL、PROCHECK和Py MOL等生物信息学软件分别对19种Mre B蛋白和16种actin蛋白进行了同源建模、质量评估、蛋白叠合比对分析;以Mre B和actin为分子指标利用MEGA6构建生物进化树分析其进化关系。研究结果表明同源建模获得的蛋白模型基本符合立体化学规则,有较高的合理性与可靠性,其中大部分属于高质量模型(落于最适区的氨基酸残基数>90%);蛋白叠合比对结果表明Mre B和actin蛋白虽为同源蛋白,氨基酸序列和结构上存在一定相似性,但一些区域仍存在明显差异。MEGA6构建的进化树基本能反映与传统分类系统一致的进化轨迹,但在某些细节方面显示出与传统分类系统相异的亲缘关系。To understand the structural dependence and evolutionary relationships between prokaryotic MreB protein and eukaryotic actin protein, Bioinformatics softwares such as SWISS-MEDOL, PROCHECK and PyMOL are applied to homologous modeling, quality evaluation and comparative analysis of protein polymerization on 19 MreB proteins and 16 actin proteins. Using MreB and Actin as molecular indexes MEGA6 is used to draw evolutionary tree to analyze their evolutionary relationships. The research shows that the protein models achieved by homologous modeling are almost in accord with the rules of stereochemistry, which has high rationality and reliability. Besides, most of them are high-quality models (The residual base of amino acid set in the optimum >90%). The result of protein polymerization comparison shows that MreB and actin proteins are homologous proteins, there is some similarity between amino acid sequence and structure, but there are still some obvious differences in some areas. The evolutionary tree built by MEGA6 relfects the revolutionary track that is consistent with the traditional classiifcation system basically, but in some speciifc aspects, it shows some genetic relationships that are different from the traditional classiifcation system.
