摘 要:膳食纤维具有重要的营养作用和保健功能,酶重量方法是测定食品中膳食纤维比较方便和经济的一种方法。参考国标GB/T 22224—2008,建立了一种测定全麦粉膳食纤维含量的方法。采用磷酸缓冲液代替MES-TRIS缓冲液,比较了磷酸缓冲液和MES-TRIS缓冲液对检测结果的影响。结果表明,使用MES-TRI缓冲液和磷酸缓冲液对测定结果影响不大,精密度均较好,方差分别为0.10和0.01。两种方法测定结果的绝对差值与平均值之比最大为4.62%,低于GB/T 22224—2008中允许的双试验误差10%。Dietary fiber has an important role in nutrition and health care function. The method of enzyme gravimetric is a convenient and economical method for the determination of dietary fiber in food. According to GB/T 22224 2008,a method for determination of total dietary fiber content of wheat flour was established. The effect of phosphate buffer and MES-TRIS buffer on the detection results was compared. The results showed that the effects of MES-TRI buffer solution and phosphate buffer on the determination results were not significant, the precision were both good,and the variance was 0.10 and 0.01 ,respectively. The maximum ratio of absolute deviation and average one by the two methods was 4.62%,which was lower than the double test error 10% in GB/T 22224 -2008.
