摘 要:目的优化唐古特青兰总黄酮提取的方法,并考察其体外抗氧化活性。方法采用单因素法与正交试验法确定了超声波辅助提取的优化条件,并探讨了总黄酮的还原能力以及对羟基自由基和超氧阴离子的清除能力。结果最佳提取工艺条件为:提取温度70℃、乙醇浓度50%、料液比1∶35;总黄酮提取物的还原能力略低于抗坏血酸,而对羟基自由基和超氧阴离子的清除率高于后者。结论超声波辅助提取唐古特青兰总黄酮具有提取率高,操作简便的特点,提取物的抗氧化活性较为明显,具有一定的实际应用价值。OBJECTIVE To study the optimum extraction conditions of total flavonoids from the Dracocephalum tanguticum Maxim and its antioxidant activity in vitro. METHODS The extraction conditions of ultrasonic method were determined by single factor and orthogonal test method. The reducing power and scavenging capacity of hydroxyl free radical and superoxide anion were investigated. RESULTS The optimum extraction conditions were as follows : Temperature of 70 ~C , ethanol concentration of 50% and solid liquid ratio was 1 : 35. The test showed that scavenging rate of total flavonoids extracts on hydroxyl radicals and peroxide anion radical were better than ascorbic acid, but the reducing power was less than latter. CONCLUSION This method is easy to operate with the high yield. The antioxidant activity of total flavonoids is obvious and worth promotion.
