摘 要:目的探究根皮苷(phloridzin)对雄性果蝇的抗衰老作用机制。方法实验以Oregon K野生型黑腹雄性果蝇为模型,将2d龄雄性果蝇随机分为4组(每组200只),即对照组和不同剂量根皮苷组(0.5、1.0和2.0 mg/ml)。定时记录果蝇死亡数目来统计果蝇的平均寿命和最高寿命,绘制果蝇寿命曲线图;测定超氧化物歧化酶(superoxide dismutase,SOD)、过氧化氢酶(catalase,CAT)和丙二醛(malondialdehyde,MDA);Real Time-PCR法测定相关基因m RNA表达水平。结果根皮苷各剂量组果蝇的平均寿命及最高寿命较对照组均显著延长(P<0.05);给予2.0mg/ml根皮苷可以显著提高Cu-Zn-SOD和CAT酶活力(P<0.01),显著降低MDA含量(P<0.01),并显著上调Cu,Zn-SOD、Mn-SOD、CAT m RNA表达水平(P<0.01),显著下调MTH(methuselah)m RNA表达水平(P<0.05)。结论根皮苷可以有效延缓果蝇衰老,并可能通过调控抗氧化基因的表达水平来实现。ObjectiveTo investigate the anti-aging effect of phloridzin in maleDrosophila melanogaster.Methods 2d-old male Drosophila were randomly divided into four groups: the control group and different doses of phloridzin treated groups (0.5, 1.0, 2.0 mg/ml) (n=200). Lifespan was calculated according to the number of dead Drosophila during each fixed period of time, and then a lifespan curve was drawn. The activities of superoxide dismutase (SOD),catalase (CAT) and content of malondialdehyde (MDA) were analyzed and the expressions of related genes were determined by real time-PCR. Results Compared with the control group, supplementation of phloridzin could significantly extend mean lifespan and maximum lifespan of Drosophila (P〈0.05). Phlorizin at 2.0 mg/ml could significantly increase the activities of SOD and CAT (P〈0.01) while MDA content decreased significantly (P〈0.01). Moreover, phloridzin at 2.0mg/ml significantly up-regulated mRNA expressions of Cu-Zn-SOD, Mn-SOD and CAT (P〈0.01) and reduced methuselah (MTH) mRNA expression (P〈0.05). ConclusionThe anti-aging effect of phlorizin on Drosophila may be related to the up-regulation of antioxidant genes expression.
