摘　　要：本实验的目的是观察大蒜素(allicin)对人结肠癌HT-29细胞凋亡和增殖的影响,并对其作用机制做初步的探讨。通过利用倒置显微镜、荧光显微镜、透射电镜、流式细胞术以及Real time PCR等方法,对大蒜素处理后的HT-29细胞的形态、存活率、细胞周期分布、细胞凋亡率、线粒体膜电位(驻追m)以及Bax/Bcl-2基因表达比例的变化进行了研究。结果显示,大蒜素可抑制HT-29细胞增殖,且呈剂量依赖性关系。大蒜素作用于HT-29细胞48 h的最佳药物浓度是6滋g/m L。在此条件下,细胞出现明显的凋亡现象。早期和晚期细胞凋亡率分别为(7.48依0.38)%和(10.07依0.78)%,驻追m显著下降(p<0.01),细胞被阻滞于S期和G2期,Bax/Bcl-2的比值显著升高(p<0.01)。因此,我们推测大蒜素可能通过调节Bax和Bcl-2两个基因的表达比例,诱导人结肠癌HT-29细胞的凋亡,进而抑制癌细胞的增殖。The objectives of this research were to investigate the effects of allicin on cell proliferation, cell cycle and cell apoptosis for human conlon cancer HT-29 cells as well as to explore the molecular mechanisms, inverted microscope, fluorescence microscope and TEM were used to observe the morphology changes in HT-29 cells after treated with allicin. The typical characteristics of cell apoptosis were studied on the treated HT-29 dells, such as the rate of apoptosis, mitochondrial membrane potential（△ψm） and cell cycle distribution of HT-29 cells. The inhibitory effect of allicin was tested on the growth of HT-29 cells by MTT. The changes of Bax/Bcl-2 expression level were measured by realtime PCR and Western blot. The results showed that allicin inhibited the growth of HT-29 cells in a dose-dependent manner. After treated with allicin 6μg/mL for 48 h, the rate of cell apoptosis went up to the highest, the level of △ψm decreased and the cell cycle were blocked in S and G2 phase.Interestingly, allicin also increased the ratio of Bax/Bcl-2 expression level. Therefore, we proposed that allicin suppress the cell growth and induce the apoptosis of human conlon cancer HT-29 cells by regulating the ratio of Bax/Bcl-2 expression level.
