摘　　要：目的研究Survivin和cyclin D1在大肠癌的表达及临床意义。方法应用Real-time PCR和Western blot分别检测大肠癌及癌旁组织中Survivin、cyclin D1 mRNA和蛋白表达情况,并探讨其临床意义。结果癌旁组织与大肠癌组织RT-PCR产物经琼脂糖凝胶电泳结果表明,Cyclin D1及survivin mRNA和蛋白质在大肠癌中高表达,而癌旁组织中不表达或低表达,两者比较,差异均有统计学意义(P<0.05)。Western blot结果表明,Survivin、cyclin D1在结直肠癌组织中均呈高表达(P<0.05)。结论 Cyclin D1和survivin的表达水平与大肠癌发生发展及恶性程度密切相关,有望成为大肠癌诊断的新靶点。Objective To study the expressions of surviving and cyclin D1 of colorectal cancer and the clinical significance. Methods Real- time PCR and western blot were used to detect surviving and cyclin D1 mRNA and protein expressions in colorectal tissue and para- carcinomatous tissue and explore their clinical significance. Results The agarose gel electrophoresis（ AGE） test results of RT- PC R in colorectal tissue and para- carcinomatous tissue showed that cyclin D1 and survivin mRN and protein expressions were high in colorectal cancer and the expressions were absent or low in the para- carcinomatous tissue,indicating the significant differences in comparison（ P〈 0. 05）. Western blot results showed that surviving and cyclin D1 were highly expressed in the colorectal cancinomatous tissue（ P 〈0. 05）. Conclusion Cyclin D1 and survivin expressions are closely related to the occurrence,development and malignancy degree of colorectal cancer and expected to be the new targets for the diagnosis of colorectal cancer.
