摘 要:针对精炼食用植物油中DNA含量低且破坏严重的问题,以CTAB(十六烷基三甲基溴化铵)法为基础进行DNA提取方法的优化,建立了可以从精炼食用植物油中有效提取DNA的方法。通过与标准方法SN/T1203-2010相比较,该方法可有效检测出内源基因tRNA Leu,但对标示有转基因成分的精炼食用植物油却无法检测到外源基因CaMV 35S,推测其原因可能为目的基因片段不够稳定,在食用植物油加工过程中被破坏,导致无法检出,在后续研究中需设计更加稳定的目的基因片段。According to the characteristics of low content and serious degradation of DNA in refined edible vegetable oils, a high efficient extraction method of DNA from refined edible vegetable oils was established based on CTAB (hexadecyl trimethyl ammonium bromide) method. Compared with the standard method SN/T 1203-2010 ,the established method could detect the endogenous gene tRNA Leu effectively, but it could not detect the exogenous gene CaMV 35S in the refined edible vegetable oils marked with genetically modified ingredient, and the possible reason was that the unstable fragment of target gene was damaged during the processing of edible vegetable oils, which showed that more stable target gene frag- ments should be designed in future research.
