摘  要:目的:筛选亚贡多糖的脱蛋白工艺。方法:采用酶法脱蛋白,应用Box-Behnken响应面法优化最佳工艺,以多糖质量分数和蛋白脱除率为评价指标,考察温度、反应时间及酶浓度对指标的影响。结果:最优工艺条件为:反应温度为56.2℃,反应时间为1.27h,酶终浓度3.7%;多糖质量分数达67.5%,蛋白脱除率为80.4%。结论:该工艺简单可行,可用于亚贡多糖的纯化。Objective To optimum conditions for deproteinization form polysaccharides of Smallanthus sonchilius. Methods Box - Behnken Design - Response surface methodology was used to optimize the process of deproteinization and the method was evaluated by the ratio of protein removing and the mass fraction of polysaccharide. Results The better conditions for the deproteinization of crude pol- ysaccharide from Smallanthus sonchilius were that the temperature was 56. 2℃, the final concentration of enzyme was 3.7% (m/v) and the reaction time was 1.27 h. Under these conditions, the content of crude polysaccharide was 67.5% and percentages of protein removing of crude polysaccharide was 80. 4%. Conclusion The method of enzyme effective method for removal of protein.
