摘  要:目的观察顺铂耐药卵巢癌细胞系COC1/DDP中多药耐药相关蛋白2(MRP2)基因的表达,探讨其反义寡脱氧核苷酸(ASODN)对COC1/DDP顺铂耐药的逆转作用。方法设计合成针对MRP2的ASODN,以200 nmol·L-1浓度转染COC1/DDP细胞,用四甲基偶氮唑蓝法观察顺铂对细胞抑制率的影响,逆转录PCR检测MRP2基因表达水平的变化。结果 COC1/DDP细胞中MRP2基因呈高表达,其mRNA相对表达值为0.420 0±0.050 0,而COC1细胞中无表达。与COC1/DDP组比较,MRP2-ASODN组MRP2基因低表达,mRNA相对表达值为0.018 7±0.023 0,耐药指数降低为4.58,差异均有统计学意义(P<0.05);SODN组MRP2表达及耐药指数改变差异均无统计学意义(P>0.05)。结论 MRP2基因高表达与COC1细胞获得顺铂耐药性有密切关系;MRP2-ASODN能有效逆转COC1/DDP细胞对顺铂的耐药性。Objective To study the expression of multi-drug resistance related protein-2(MRP2)of COC1/DDP cell line,and the reversion of cisplatin resistance by antisense oligodeoxynucleotide( ASODN). Methods The ASODN targeting MRP2 was designed and synthesized. The ASODN targeting MRP2 with the concentration of 200 nmol·L-1 was transferred into COC1/DDP cells by lipofectin. The inhibition rate of transferred cells to cisplatin was detected with MTT assay. The altered expression of MRP2 was detected by reverse transcription PCR. Results MRP2 were overexpressed in cisplatin resistant cell line of COC1/DDP compared with COC1 cell line. Compared with the COC1/DDP cell group,MRP2 expression in the MRP2-ASODN transfected cell group was significantly de-creased to 0.018 7 ±0.023 0,and the resistance index was 4. 58(P〈0. 05);there were no difference in the MRP2 expression and resistance index between the COC1/DDP cell group and the SODN group(P〉0. 05). Con-clusion The overexpression of MRP2 may play an important role in the induction of resistance of COC1/DDP cells to cisplatin;MRP2-ASODN can effectively reverse the resistance of COC1/DDP cells to cisplatin and increase their drug sensitivity in a sequence specific manner.
