摘  要:实验采用木聚糖为唯一碳源,利用平板透明圈筛选和摇瓶培养基发酵相结合的方法,筛选得到一株具有木聚糖降解效果的菌株,记为2a1,经形态学和分子生物学鉴定为曲霉属(Aspergillus)。通过液体摇瓶发酵对该株菌发酵产木聚糖酶所需的碳源、氮源、起始pH值、培养温度、培养时间等条件进行研究,从实验结果可以看到上述因素均对产酶量有一定的影响。在单因素的基础上,设计一个3因素3水平的正交试验,根据证交试验结果确定这株菌株的最佳产酶条件为:产酶培养基中木聚糖添加量为6g·L-1,起始pH值为4.5,30℃,150r·min-1恒温振荡箱培养84h;其中起始pH值的影响最为显著。In this experiment,a strain,having the ability of degrading xylan,was screened out adopting the method of flat transparent circle screening method combined with shake flask method and with xylan as sole carbon source. The strain was denoted as strain 2a1,and was identified morphologically and molecular biologically as aspergillus. Its xylanase fermentation conditions,namely of carbon source,nitrogen,initial p H,temperature,incubation time,were investigated by shaking flask fermentation. The result showed that all above factors had impact on the amount of xylanase produced. A orthogonal test obtained an optimum xylanase producing condition: xylan concentration of 6 g·L- 1,initial p H of 4. 5,30℃,incubating in 150 r·min- 1oscillation incubator for 84 h. The initial p H had the most influence on the xylanase producing fermentation.
