摘  要:目的 探讨绿茶多酚(GTPs)对高脂膳食大鼠心肌窖蛋白-1(caveolin-1,cav-1)和热休克蛋白(heat shock protein 90,HSP90)表达的影响及可能的心脏保护作用。方法 体重80~100g雄性wistar大鼠40只,随机分为正常对照组、高脂对照组、绿茶多酚干预组(两个剂量组)4组,每组10只。对照组分别自由摄食标准饲料和高脂饲料,绿茶多酚干预组在自由摄食高脂膳食的同时给予不同浓度的绿茶多酚水溶液(0.8 g/L和1.6 g/L)。35 w后将大鼠断头处死并收集全血及心脏组织;采用试剂盒测定血清甘油三酯,胆固醇、血糖、胰岛素和胰岛素抵抗(HOMA-IR)指数;采用RT-PCR法和Western-blotting法分别测定HSP90和cav-1的m RNA及蛋白表达水平。结果 与正常对照组比较,高脂组体质量、血清甘油三酯、总胆固醇和血糖等指标均显著升高(P〈0.05),茶多酚干预后有所改善(P〈0.05);高脂组HSP90 m RNA及蛋白水平降低,caveolin-1 m RNA及蛋白水平升高(P〈0.05),绿茶多酚干预后HSP90表达升高,cav-1表达降低。结论 绿茶多酚可能通过上调HSP90和下调caveolin-1转录及蛋白表达水平,在高脂膳食条件下发挥对心肌的保护作用。Objective To explore the possible cardioprotective effects of green tea polyphenols(GTPs) on the expressions of caveolin-l(cav-1) and heat shock protein 90 (HSP90) in rats fed a high fat diet. Methods Forty male Wistar rats were randomly divided into four groups of equal size: the control group (CON), fed a standard chow; the high fat group (HF), fed a high fat diet; two GTPs treated groups, fed a high fat diet and different concentrations of green tea polyphenols solution (0.8 g/L and 1.6 g/L). Deionized water was given to CON and HF group. After thirty-five weeks, the blood and heart tissue were collected. The levels of triglyceride, cholesterol, glucose, insulin, and insulin resistance (HOMA-IR) index in serum were measured. The mRNA and protein expressions of HSP90 and caveolin-1 in heart were measured by real-time polymerase chain reaction (real-time PCR) and Western blotting, respectively. Results The body weight and levels of serum triglyceride, cholesterol, glucose, insulin and HOMA-IR in HF group were significantly higher (P〈0.05) than those in control group. Additionally, the HF group showed lower mRNA and protein expression levels of HSP90 but higher levels of eaveolin-1 than those in control group (P〈0.05) However, in GTP groups, the body weight and the levels of serum glucose and HOMA-IR significantly decreased compared with those in HF group. Moreover, compared with HF group, the mRNA and protein expression of HSP90 in GTP groups were higher, while those of Cav-1 significantly decreased in GTP groups. Conclusion GTPs can up-regulate the expression of HSP90 and down-regulate that of Cav-1, and those may contribute to its the cardioprotective effects.
