摘 要:[目的]在恶劣环境下,摸索酵母菌的生活方式、生长条件,为完善贵州酵母菌资源收集、探究酵母菌的抗逆性及共享微生物菌种资源奠定基础。[方法]参照柠檬形克勒克酵母(Kloecken apis)的生长特性,以培养特性、形态学特征及生理生化特性为基础,按照Lodder分类方法对XGF-14菌株进行初步鉴定。[结果]从淤泥中分离出一株菌落为红色的酵母菌,接种该菌于葡萄糖-酵母膏-蛋白胨琼脂培养基上,在28℃培养条件下观察发现:菌落前期粉红色、后期为桔红色;菌落表面光滑、湿润、有光泽、边缘整齐、圆形(直径约1 mm)、稍有粘性、中央突起。该酵母菌对大部分糖类起不了同化作用,但在发酵葡萄糖时有红色菌环产生。单端或多端芽殖,形成假菌丝。[结论]根据酵母菌的形态特征、生理生化特性能初步鉴定XGF-14为红色冬孢酵母属(Rhodosporidium)近梨形红冬孢酵母(Rhodosporidium dacryoidum)。由于该酵母菌落颜色的特殊性,为天然色素的发展提供理论参考。[Objective] In this study,groping for yeast's life and growth conditions under the worse environment to lay the foundation for collection of yeast resources,exploring the growth of yeast adversity and sharing microorganism resources in Guizhou.[Method]Compared with growth characteristics of Kloecken apis, the method of morphological, physiological and biochemical characteristics of XGF-14 strains were adopted to identify via Lodder technique.[Result]The red yeast was isolated from the sludge, this yeast was cultivated on the glucose-yeast extract-peptone agar medium by 28 ~C.This process, we observed that the early colonies pink, the latter part of the orange-red, the surface smooth, moist, shiny, neat edges, round (diameter 1 mm or so),slightly viscous,central protuberance.The yeast couldn't play the assimilation role on most sugar,while it produced red collarium under fermenting glucose.Single side or multiterminal budding,pseudohyphae. [Conclusion]We initially identified a red yeast as Rhodosporidium dacryoidum based on the morphological characteristics and physiological and biochemical characteristics.In the process of cultivation ,Rhodosporidium dacryoidum had colony color varies from time to present different colony color to provide a theoretical basis for the function of the yeast.
