摘 要:目的优化共附生链格孢发酵条件,并分析其产生的抗菌物质活性影响因素。方法采用单因素实验和田口试验设计方法对共附生链格孢P11发酵培养基及发酵条件进行优化,同时对可能影响发酵液中抗菌物质稳定性的因素进行研究。对发酵液的石油醚、乙酸乙酯和正丁醇的萃取物进行抗菌活性测试,了解抗菌物质的极性及疏水特性。结果发酵培养基的最佳组合为葡萄糖10g,胰蛋白胨25g,K2HPO4·3H2O 0.6g,KCl 1g,海水1L,pH值6.5。链格孢P11菌株产生的抗菌物质随着热处理时间延长抗菌活性降低,但对pH值(1~13)、白炽灯和紫外线(波长365nm)表现出较好的稳定性。正丁醇提取物抗菌活性最高。结论通过优化实验确定了菌株P11最优发酵培养基及发酵条件,其产生的抗菌物质有较好的稳定性,有较大极性和水溶性。这为具有生物活性的次级代谢产物的分离奠定了基础。Objective To optimize the fermentation conditions of Endophytic Alternaria sp.and analyze the influencing factors that impact its antibacterial substance. Methods Single-factor experiment and DOE tests were used to optimize the fermentation medium of Endophytic Alternaria sp.P11 and its fermentation conditions,and research the factors that may impact the stability of antibiotic substances in fermentation broth.An antibacterial activity test was done for the extractant of ligarine,acetic ether,and n-butyl alcohol in the fermentation broth to investigate the polarity and hydrophobic property of the substances. Results The best combination of fermentation medium was glucose(10g),tryptone(25g),K2HPO4(0.6g),KCl(1g),seawater(1L),and pH 6.5.Antibacterial activity of antibiotic substances produced by strain Alternaria sp.P11 decreased with the heat treatment time prolonged,but showed good stability for pH(1-13),incandescent lamp and ultraviolet rays(wave length365nm).The antibacterial activity of butanol extract was the highest. Conclusion Through optimization experiment,the best fermentation medium and conditions for the P11 strains are determined,the antibacterial substances produced have great polarity and water solubility,which lays the foundation for isolation of secondary metabolites of that with biological activity.
