摘 要:目的通过建立微分方程模型,探讨鼻咽癌血浆Epstein-Barr病毒-DNA(EBV-DNA)含量在放疗中的动力学变化。方法选取2013年1月至2014年12月10例接受单纯放疗的鼻咽癌患者,采用荧光定量PCR检测其血浆EBV-DNA的动态表达水平。结果微分方程模型能很好地拟合血浆EBV-DNA的实际观测数据。模型参数与治疗中EBV-DNA的清除动力学特征有很好的相关性,且可能对患者的临床表型有一定的指示作用。结论微分方程模型参数可能提示肿瘤的生物学特性,但其意义价值尚需进一步验证。Objective To develop a differential equation kinetic model,and explore the dynamics of plasma Epstein-Barr virus( EBV)-DNA concentration for nasopharyngeal cancer( NPC) patients undergoing radiation therapy. Methods Ten NPC patients treated with intensity modulated radiotherapy from January 2013 to December 2014 were involved in this study. The content of EBVDNA in the plasma samples were detected by using fluorescent quantitative PCR method. Results Fitting of the model to the data of plasma EBV-DNA levels allowed determination of the different parameters describing the radiosensitivity,dead-cell resolving and necrotic cell fraction. These parameters were correlated with patterns of the dynamics of plasma EBV-DNA level during radiotherapy,and it may be indicative of the clinical phenotype. Conclusion The model parameters may have a potential to predict biological characters of tumor,though further validation in a larger pool of samples is warranted.
