摘 要:[目的]建立白、红花生衣多酚物质含量的分析方法并优化其提取技术。[方法]通过光谱特征分析白、红2种花生衣多酚物质主要组成,并建立多酚物质含量分析方法。以60%乙醇为提取剂,通过多次单因素试验,对超声辅助与微波辅助提取的最佳条件进行了优化,比较超声辅助、微波辅助及振荡提取花生衣多酚含量的差异。[结果]通过花生衣多酚物质的紫外可见光谱特征,说明这2种花生衣多酚物质以原花青素和儿茶素等单体为主,从而建立了以原花青素和儿茶素为对照的紫外法分析其多酚物质含量的方法。试验表明,经超声辅助提取多酚物质含量比微波辅助提取略高,二者比振荡提取都要高一些。[结论]花生衣多酚物质含量分析采用紫外法符合其光谱吸收特征,结果可靠。[ Objective ] To establish analysis way of polyphenol in white and red peanut skin and optimize its extraction method. [ Method ] According to the spectrum feature of polyphenol in red and white peanut skin, the major components of it was analyzed, and the analysis way of polyphenol in red and white peanut skin was established. With 60% ethanol as extracting agent, and by se- ties of single-factor tests, the best conditions of ultrasonic-assisted and microwave-assisted extraction of polyphenols were obtained. The content of polyphenol in peanut skin extracted by three methods of ultrasonic-assisted, microwave-assisted and oscillation was compared too. [ Result] Through analyzing spectral characteristics of polyphenol in red and white peanut skin, it showed that polyphenol in red and white peanut skins mainly contains proanthocyanidin and catechin, etc, monomers. So the UV analysis way of polyphenol in red and white peanut skin, which used proanthocyanidin and catechin as standard sample, was established. The results showed that ultrasonic-assisted extraction was a little more effective than microwave-assisted extraction and Oscillation extraction. [ Conclusion] The content of polyphenols in peanut skin was ana- lyzed by UV method, which was consistent with the absorption characteristics and the results were reliable.
