摘 要:目的探讨虾青素对过氧化氢诱导星形胶质细胞氧化损伤的保护作用。方法星形胶质细胞传代培养后,分别加入不同浓度(1.0×10-7mol·L-1、1.0×10-4mol·L-1)虾青素预处理12 h后,加入100μmol·L-1H2O2继续孵育24 h,MTT比色法检测虾青素对H2O2诱导的星形胶质细胞活力的影响,流式细胞仪检测细胞凋亡率,比色法检测凋亡相关因子caspses-3及caspase-9的表达。结果虾青素孵育24 h后细胞活性分别为101.44%±1.58%,98.76%±1.53%,101.33%±2.28%,98.67%±1.62%,与对照组(98.33%±0.93%)比较差异无统计学意义(P>0.05);模型组星形胶质细胞经氧化损伤处理后,细胞活性明显下降(32.55%±3.81%),用1.0×10-7mol·L-1及1.0×10-4mol·L-1虾青素处理后,星形胶质细胞活性分别提高到53.67%±4.41%和69.33%±3.01%,与模型组比较差异具有统计学意义(P<0.05)。流式细胞计数结果显示:正常对照组星形胶质细胞平均凋亡率为4.11%±1.54%,经H2O2处理后,模型组星形胶质细胞凋亡平均率为39.55%±4.28%,应用虾青素后,高剂量组星形胶质细胞平均凋亡率为16.19%±3.61%,与模型组比较,差异具有统计学意义(P<0.01)。此外虾青素还减少了H2O2所致星形胶质细胞内caspses-3及caspase-9的表达,并且,伴随虾青素作用时间的延长其表达量呈下降趋势。结论虾青素可以明显抑制过氧化氢诱导的星形胶质细胞的凋亡,从而为其用于治疗中枢神经疾患提供实验依据。Objective To investigate the protective effect of astaxanthin against hydrogen peroxide( H2O2)-induced oxidative damage to astrocytes. Methods Subcultured astrocytes incubated with different concentrations( 1. 0 × 10- 7mol·L- 1、1. 0 × 10- 4mol·L- 1) of astaxanthin for 12 h and 100 μmol·L- 1H2O2 for 24 h,the impact of astaxanthin on H2O2-induced cell viability was detected by MTT assay,rate of apoptosis was detected by flow cytometry and apoptosis-related factors of caspses-3 and caspase-9 were detected by colorimetric method. Results Cell viability after 24 h incubation of astaxanthin was 101. 44% ± 1. 58%,98. 76% ±1. 53%,101. 33% ± 2. 28%,and 98. 67% ± 1. 62%,respectively,there was no statistically significant difference compared with the control group( 98. 33% ± 0. 93%); Cell viability decreased significantly( 32. 55% ± 3. 81%) in H2O2-treated group,but it was improved after1. 0 × 10- 7mol · L- 1( 53. 67% ± 4. 41%) and 1. 0 × 10- 4mol · L- 1( 69. 33% ± 3. 01%)astaxanthin treatment,there was statistically significant difference compared with the model group( P 0. 05). Flow cytometry results showed that the apoptosis rate in normal control group of astrocytes was 4. 11% ± 1. 54% and 39. 55% ± 4. 28% in model group,however,after the application of astaxanthin,cell apoptosis in astaxanthin high dose group decreased to16. 19% ± 3. 61%,the difference was statistically significant compared with the model group( P 0. 01). Besides,the astaxanthin treatment also reduced H2O2-induced expression of caspses-3 and caspase-9 in astrocytes. Conclusion Astaxanthin can inhibit apoptosis induced by hydrogen peroxide over astrocytes,which provides experimental evidence for treatment of central nervous disorders.
