摘  要:根据植物花青素合成途径关键酶基因序列设计特异性引物,利用RT-PCR技术,在转录水平上检测这些基因在彩色马铃薯不同颜色块茎中的表达情况。从18个基因中筛选到类黄酮3’5’-羟化酶基因、二氢黄酮醇还原酶基因、类黄酮3-O-葡萄糖基转移酶基因、类黄酮5-O-葡萄糖基转移酶基因、甲基转移酶基因、谷胱甘肽转移酶基因、调节基因AN2和AN1等8个基因在不同颜色薯肉中表达有差异,说明这些基因的表达可能与块茎肉色相关。研究检测的基因中有8个在马铃薯中未见报道,本研究根据矮牵牛的基因序列,寻找马铃薯的同源序列,经检测这些同源序列在彩色马铃薯块茎中都有表达产物,表明这些同源序列可能是目的基因,研究结果为今后在马铃薯上分离克隆花青素合成途径关键酶基因奠定基础。Special primer designs were based on the sequences of key genes in the anthocyanin biosynthetic pathway in higher plants. The key genes expressions were studied in different color tubers by RT-PCR technique. There were five genes flavonoid 3 ',5'-hyroxylase,dihydroflavonol 4-reductase,flavonoid 3-O-glucosyltransferase,flanonoid 5-O-glucosyltransferase,AN1,methyltransferase,glutathione S-transferase,AN2 and AN1 genes,which were related to potato tuber color expression. There were 8 key genes involved in the anthocyanin synthesis which had not been reported in potato. Our research found their homologous sequence in potato based on the petunia gene sequences. The expression products of these homologous sequences were detected in potato. The results could be very useful for the genetic clones generated in near future.
