摘 要:目的:初步研究非洲马铃薯水提物改善骨骼肌糖脂代谢作用机制。方法:40只雄性SD大鼠,取10只大鼠作为正常组,取30只大鼠高脂喂养1个月加一次性腹腔注射STZ造模,造模成功后分为模型组、吡格列酮组、南非马铃薯组。分组灌胃5周。每周测体质量、空腹血糖(Glu);实验结束后取血清测Glu、甘油三酯(TG)、总胆固醇(TC)、高密度脂蛋白(HDL)和低密度脂蛋白(LDL)含量;取骨骼肌测超氧化物歧化酶(SOD)、丙二醇(MDA)和细胞凋亡,测Mn SOD、GLUT4蛋白及m RNA表达情况。结果:与正常组比较,模型组大鼠Glu、TG、LDL升高(P<0.01),大鼠骨骼肌抗氧化酶SOD减少(P<0.01),线粒体抗氧化酶Mn SOD减少(P<0.01),氧化产物MDA含量增加(P<0.01),GLUT4表达受抑制(P<0.01),且TUNEL显示骨骼肌细胞凋亡增多。南非马铃薯具有改善上述异常变化的作用。结论:南非马铃薯可能通过抗氧化应激提高骨骼肌Mn SOD含量,促进GLUT4表达改善骨骼肌的糖脂代谢。Objective: To investigate the mechanism of African potato(AP) aqueous extract improving the glucolipid metabolism in skeletal muscles. Methods: 40 male SD rats were randomly divided into 4 groups: control group, model group, pioglitazone hydrochloride group and AP group, each group for 10 rats. All groups except for the control group were given high fat diet for a month and intraperitoneal injected of STZ to establish the diabetes mellitus models. After modeling, two treatment groups and the model group were given the medicine and water by gavage for 5 weeks, respectively. Body weight and fast blood glucose were measured every week. The contents of Glu, TG, TC, HDL and LDL in blood serum were detected after the rats were sacrii ced. The contents of SOD, MDA and apoptosis were detected in skeletal muscle. And, the protein and m RNA expressions of Mn SOD, GLUT4 were detected in skeletal muscle. Results: Compared with the control group, the Glu, TG, LDL in the model group increased(P〈0.01). The antioxidase SOD and Mn SOD of skeletal muscle in the model group declined(P〈0.01), the content of oxidative product MDA increased and the expression of GLUT4 was inhibited in the model group(P〈0.01). The TUNEL staining showed that the apoptosis of skeletal muscle cell increased. The AP could improve these changes above. Conclusion: It is possible that AP resisted oxidative stress to increase the content of Mn SOD and the expression of GLUT4, which can improve the lucolipid metabolism in skeletal muscles.
