水产品中霍乱弧菌的检测方法研究

摘　　要：目的研究出一种快速、敏感、高效、实用的水产品中霍乱弧菌检测方法,以便更好地监测外环境和水产品中霍乱弧菌污染状况,及时有效地防控霍乱疫情。方法按照《全国霍乱监测方案》,配制含霍乱弧菌的模拟水产品涂抹样标本,在传统碱性蛋白胨水中加入1μg/ml的萘啶酸,并在传统庆大分离平板中减少庆大霉素含量至250 IU/L,再加入1μg/ml的萘啶酸,分别用胶体金法、上转发光法、实时荧光PCR法、分离平板法,检测不同浓度霍乱弧菌的水产品涂抹样增菌后霍乱弧菌的检出效果。结果用改良法增菌后,胶体金法、上转发光法、实时荧光PCR法检测霍乱弧菌,阳性检出率普遍提高,再用改良法分离培养,霍乱弧菌分离率大大提高。结论在进行水产品中霍乱弧菌监测时,在增菌液中加入1μg/ml的萘啶酸,在庆大培养基中减少庆大霉素含量至250 IU/L,再加入1μg/ml的萘啶酸,可以抑制杂菌,促进目标菌生长,提高霍乱弧菌的检测效果。Objective To study a rapid, sensitive, efficient and practical method for detection of Vibrio cholera in aquatic prod- ucts, in order to better monitor the environmental and aquatic products in Vibrio cholera contamination status for timely and ef- fective prevention and control of cholera epidemic. Methods According to the National Plan for Cholera Surveillance, to pre- pare smear specimens of simulated water products containing Vibrio cholerae, adding 1 μg/ml nalidixic acid to traditional alka- line peptone water, and in the traditional qing large separation plate to reduce the gentamicin concentration to 250 IU/L, and then 1 μg/ml nalidixic acid more, with colloidal gold method, turn luminescence method, real -time fluorescence PCR meth- od, separation plate method, to detect detection effect of Vibrio cholerae in different concentrations of Vibrio cholerae in aquatic products smear samples after increasing bacteria. Results After using the improved method to increase the fungus, colloidal gold method, the forward light method, real - time fluorescent PCR method were used to detect Vibrio cholerae. With the posi- tive detection rate rising generally, cultivation by using the improved method, separation rate of Vibrio cholera greatly increased. Conclusion In the monitoring of Vibrio cholera in aquatic products, to add 1 μg/ml nalidixic acid in the enrichment solution, and the content of gentamicin was reduced to 250 IU/L, adding 1 μg/ml nalidixic acid again in it, which can inhibit the growth of the others bacteria and promote the growth of Vibrio cholera, and can improve the detection effect of Vibrio cholera.
