摘 要:目的研究醒脑丸对血管性痴呆(VD)大鼠脑组织中乳酸(LD)、谷氨酸(Glu)含量的影响。方法先将60只SD大鼠随机分为正常组8只和造模组52只,造模组采用结扎双侧颈总动脉(2-VO)法制作VD模型,然后将造模成功VD大鼠随机分为模型组、脑复康组、醒脑丸低剂量组、醒脑丸中剂量组、醒脑丸高剂量组。造模35 d后,脑复康组开始给予脑复康84 mg/kg灌胃,醒脑丸低、中、高剂量组分别给予醒脑丸水溶液0.21 g/kg、0.42 g/kg、0.84 g/kg灌胃,正常组和模型组给予等量生理盐水灌胃,均每天1次,持续14 d;末次灌胃后当天进行大鼠行为实验,采用比色法测定大鼠脑组织中LD、Glu含量。结果模型组大鼠学习能力明显低于正常组(P<0.05),各给药组大鼠的学习能力明显高于模型组(P均<0.05)。模型组大鼠脑组织中LD、Glu含量均明显高于正常组(P均<0.05),各给药组大鼠脑组织中LD、Glu含量明显低于模型组(P均<0.05)。结论醒脑丸能够改善VD大鼠的学习记忆能力,其作用可能是通过降低脑组织中Glu及LD含量实现的。Objective It is to investigate the influence of Xingnao Pill on glutamic acid and lactic acid content of cerebral tissue in vascular dementia rat. Methods Sity rats were randomly selected 8 cases into normal group,the others were duplicated models of vascular dementia rats with the operation of ligating bilateral carotids,and then divided into model group,Naofu Kang group,low-dose Xingnao Pill group,medium-dose Xingnao Pill group,high-dose Xingnao Pill group. At the thirty-fifth day after operation,the animals were treated with Naofu Kang 84 mg / kg gavage in Naofu Kang group,and with Xingnao Pill water solution 0. 21,0. 42 and 0. 84 g / kg gavage in low-,medium-and high-dose Xingnao Pill group,same amount of normal saline were gavage in normal group and model group,once a day for fourteen days. At the last day of treatment,the behavior experiment of rats were executed,then the glutamic acid and lactic acid content of cerebral tissue in rats were determined. Results The learning and memory ability of rats in the model group was obviously decreased than that in the normal group( P 〈0. 01),the Learning and memory ability of rats in the groups with drug were improved than that in the modle group( all P 〈0. 05). The levels of glutamic acid and lactic acid in cerebral tissue of rat in model group was markedly higher than that in the blank group( P 〈 0. 01),and that in each medical group was lower than that in model group( all P 〈 0. 05). Conclusion Xingnao Pill could treat vascular dementia by reducing glutamic acid and lactic acid content in cerebral tissue.
