摘  要:目的探讨丁基苯酞对缺血性脑卒中大鼠工作记忆和皮质区Notch1表达的影响。方法 SD大鼠分为假手术组(对照组)、模型组、丁基苯酞高、低剂量组。参照改良的Pulsineli 4血管阻断建立大鼠缺血性脑卒中模型;HE染色观察各脑组织病理学形态的变化、免疫组化法检测Notch1的表达;八臂迷宫法和Morris水迷宫测试动物工作记忆情况。结果与对照组比较,脑缺血后皮质区神经元死亡数量、Notch1水平增加;八臂迷宫测试的动物工作记忆错误次数增多、水迷宫测试的穿台次数明显减少,差异有统计学意义(P<0.05)。与模型组比较,丁基苯酞组中大鼠脑缺血后海马区神经元死亡数量降低、Notch1水平增加;八臂迷宫测试的动物工作记忆错误次数减少、水迷宫测试的穿台次数明显增多,差异有统计学意义(P<0.05);上述变化在高剂量丁基苯酞组最为显著(P<0.05)。结论丁基苯酞可增加Notch1的表达、改善缺血性脑卒中大鼠工作记忆能力。Objective To investigate the effect of dl-3n-butyphthalide(NBP)on working-memory function and Notch1 expression in cortex in rats with ischemic stroke.Methods Healthy adult male SD rats were randomly divided into control group,model group,low dose of NBP group and high dose of NBP group.Ischemic stoke model was formed by improved four-vessel occlusion method.Morphological changes in hippocampal region were observed by HE staining.The expression of Notch1 was detected by immunohistochemistry.Working- memory function was evaluated by Eight-arm maze and Morris water maze daily.Results Compared with control group,necrosis neurons and Notch1 expression enhanced;The working memory error frequencies increased and frequency of crossing the platform decreased significantly;Compared with model group,necrosis neurons decreased while Notch1 expression increased further;The working memory error frequencies decreased and the frequency of crossing the platform increased significantly in NBP groups.What the above mentioned index changed more significantly in high dose of highdose NBP group.Conclusion NBP can improve learning-memory function after ischemic stoke,and the molecular mechanisms is related to enhance the Notch1 expression.
