摘  要:从雉鸡肠道分离得到1株生长良好的乳酸菌DCe13,菌株革兰氏阳性、接触酶阴性.经系统发育和recA基因特定片段分析,结合菌落形态、细胞形态、生化反应试验,确定菌株DCe13为植物乳杆菌.菌株DCe13培养36 h,乳酸含量9.08 μg/mL,而pH值下降为4.7.菌株具有酸耐受性、胆盐耐受性和一定的胆固醇清除能力.菌株在pH2.0培养2h后仍能达到7.9×10^4CFU/mL;5 h内,加胆盐(0.3%~0.4%)对菌株生长量(OD值)没有影响.胆固醇去除量与菌体的生长呈正相关,培养48 h,单位细胞于重能清除最大的胆固醇量为22.71.μg/mg.另外,对比试验中生长细胞单位干重去除的胆固醇量达到10.50 μg/mg,但热杀死和休眠细胞能去除很少的胆固醇,分别是3.59 μg/mg和8.26 μg/mg.菌株DCe13去除胆固醇可能的机理是菌体对胆固醇的吸附及菌体在生长过程中对胆固醇的吸收利用.研究表明该菌株具有添加到食品中降低血液胆固醇的潜能.A Gram ( + ), non-motile, facuhatively anaerobic, short-rod shaped bacterium, designated strain DCel3, was isolated from the pheasant intestinal. The 16S rRNA gene sequence and recA gene fragment analysis showed that strain DCel3 belonged to Lactobacillus plantarum. As DCel3 growing in MRS media, the amount of lactic acid in the fermented broth was increasing, while pH value falling continually. After 24 hours, lactic acid concentra- tion reached 9.08 p,g/mL. Strain DCel3 showed acid resistance, bile salt tolerance and the cholesterol-reducing abil- ity. The results indicated that strain DCel3 survived at pH2 after 2 h, and the living bacterial number could reach 7.9 × 104 CFU/mL. During 5 h of incubation, strain DCel3 growing in MRS media complemented with 0.3% ~ 0.4% bile salt was impacted. Cholesterol removal was positively related to growth of cultures and reached its maxi- mum (22.71 μg/rag of dry weight) at 48 hours. The cholesterol assimilation by growing cells was 10.50 μg/mg of dry weight, which was significantly higher compared with the resting and dead cells. While cholesterol removed by dead and resting cells was 3.59 μg/mg and 8.26 μg/mg of dry weight, respectively. These evidences indicated cho- lesterol removal by adsorption or absorption. This study confirmed the potential of DCel3 strains for application in functional food formulations especially when cholesterol reduction in food was needed.
