摘 要:目的观察三种浓香型白酒对小鼠记忆功能的影响,分析不同白酒致机体记忆损伤的差异,探讨记忆功能损伤可能的分子机制。方法80只雄性昆明种小鼠随机分为4组(1组为对照组,2-4组为酒样组),灌胃剂量从12ml/(kg·d)开始,每3天增加1ml/(kg·d)直到16ml/(kg·d),连续灌胃6周,Morris水迷宫试验观察小鼠记忆功能情况,ELISA方法检测血清神经烯醇化酶(NSE)的水平,实时荧光定量PCR(RT—PCR)方法检测小鼠海马区环磷酸腺苷应答元件结合蛋白(CREB)及N-甲基-D-天冬氨酸受体(NMDAR)亚基NR1、NR2B的mRNA的表达水平,对比组间差异。结果与对照组相比,3个酒样组的水迷宫试验潜伏期延长(P〈0.05),组间两两比较,①号酒样组和③号酒样组的潜伏期短于②号酒样组p〈0.05),①、③号酒样组之间无统计学差异;海马区NR1、NR2BmRNA表达量较对照组均有上调(P〈0.05),且上调程度由高到低次序同为②号、③号、①号酒样组(P〈0.05);血清NSE水平及海马区CREB基因mRNA表达量在对照组与酒样组之间无统计学差异。结论灌胃6周后,三种浓香型白酒对小鼠的学习记忆功能均有不同程度的损伤,提示这种损伤可能与白酒种类相关,而造成损伤的机制可能与海马区NR1、NR2B基因mRNA表达上调有关。Objective To investigate the effects of three kinds of Luzhou-flavor liquor on memory function of mice and the difference among them, and discuss the possible molecular mechanism of the loss of memory function. Methods 80 male Kunming mice were randomly divided into four groups: blank control group, wine sample group ①, wine sample group ② and wine sample group ③. Control group and three wine sample groups were administered respectively with water and Luzhou-flavor liquor for 6 weeks, from 12ml/kg·d to 16 ml/kg·d by increasing lml/kg'd every three days. Then Morris Water Maze was used to observe the changes of memory ability. ELISA was done to detect the changes of the serum neuro-specific enolase (NSE), and the expression of cAMP-responsive element binding protein (CREB), NMDAR1 (NR1), NMDAR2B (NR2B) mRNA in the hippocampus were also detected by Real-time PCR (RT-PCR). Results The Morris Water Maze demonstrated that three sample groups' latency were prolonged compared with the controls (P〈0.05), and the wine sample group ① and ② were shorter than wine sample group ② (P〈0.05), but there was no significant difference between wine sample group ①and ②; Three sample groups' expression of NR1 and NR2B mRNA increased significantly (P〈0.05) in the hippocampus compared with the control group, and their increase level were both ① 〉 ② 〉 ③ in NR1 and NR2B (P〈0.05); Meanwhile, no significant alteration in serum NSE and hippocampus CREB mRNA were found between groups. Conclusion After 6 weeks of administration, all three kinds of Luzhou-flavor liquor could lead to diverse extent of memory impairment on mice, possibly associated with the specific type of wine. The memory damage of may be associated with the enhanced expression of NR1, NR2B genes and mRNA.
