Hepatoprotective Effect and Antioxidant Activity of Crude Polysaccharide from Calvatia gigantea Mycelium Grown in Submerged Culture

摘  要:以雌性小白鼠为研究对象,采用CCI,诱导的小鼠肝损伤模型,测定了大秃马勃(Calvatiagigantean)液体发酵菌丝粗多糖(CPSM)对小鼠体内免疫相关器官的相对重量、小鼠血清中的天门冬氨酸氨基转移酶(AST)和丙氨酸氨基转移酶(ALT)活性的影响;结果表明,经CCI。诱导小鼠的肝脏、脾脏和胸腺指数上升,AST和ALT活性剧烈升高;小鼠预先灌胃CPSM能缓和这些器官指数和酶活的变化,其中高剂量组(20mg/100gBW)与灌胃水飞蓟素的阳性对照结果相当。以BHT为阳性对照,检测CPSM的体外抗氧化活性表明,CPSM对DPPH自由基、羟自由基和超氧阴离子的清除能力在实验范围与用量呈正相关关系,在100μg/mL时,对DPPH、OH和O2-的清除率分别为32%、56%和48%。The protective effects of crude polysaccharide from Calvatia gigantea mycelium grown in submerged culture (CPSM) against carbon tetrachloride (CC14)-induced chronic hepatitis in female mice are described. Mice were given 0.1% CC14 , [1 mL/100 g body weight (BW)] following oral administration of CPSM daily for 15 days, and spleen, liver and thymus weights, and serum aspartate amino transferase (AST) and alanine aminotransferase (ALT) levels, were determined 16 hours later. CC14 injection increased the liver, spleen and thymus parameters by between 38% to 57%, and serum AST and ALT levels activities by 8.86 and 1.92 times, respectively. These increases were offset by prior administration of CPSM, especially in mice fed 20 mg/100 g BW CPSM. CPSM also exhibited dose-dependent DPPH, OH and 02- scavenging activities; CPSM (100 μg/mL) scavenging rates for DPPH, OH and O2- were 32%, 56% and 48%, respectively.
