摘  要:为探索金丝小枣低聚糖提取工艺及其生物活性,开发利用金丝小枣的营养功能价值,本文以酸解结合酶解的方法对金丝小枣多糖进行降解,利用正交法优化制备工艺。以3-氨基-9-乙基咔唑作为衍生剂对所制备的低聚糖进行HPLC柱前衍生,分析其单糖组成,并对其抑制食物腐败菌的活性及抗氧化性进行体外实验研究。结果表明,以0.5 mol/L的三氟乙酸,80℃水解小枣多糖6 h;对所得沉淀以0.05 mg/m L果胶酶在45℃下酶解40 min,可较高效率获得金丝小枣低聚糖,其得率为:24.87%±0.63%。单糖组分分析最佳色谱条件为:流动相28∶52,乙腈-水(0.1 mol/L醋酸铵缓冲液,p H4.3);流速0.8 m L/min;UV 254 nm。所制备的小枣低聚糖经测定具有抑菌活性,对大肠杆菌、枯草芽孢杆菌、金黄色葡萄球菌的最小抑菌浓度分别为0.195、0.781、0.391 mg/m L,且具有一定的抗氧化能力,在其浓度为25 mg/m L时,总抗氧化能力为(4.69±0.14)U,DPPH·清除率为57.71%±0.51%,·OH清除率为35.24%±1.08%。说明金丝小枣具有功能营养开发利用的价值。To explore the extraction process and bioactivities of oligosaccharides from Zizyphus Jujuba cv.Jinsixiaozao as well as utilization of its nutrition value,the methods of acidolysis and enzymolysis were employed to degrade polysaccharides in Jinsixiaozao,and the preparation process was optimized orthogonally.The composition of monosaccharide was analyzed by high performance liquid chromatography after precolumn derivation of oligosaccharides using 3- amino-9- ethylcarbazole as the derivating agent.In vitro assays were also conducted to investigate the antioxidant activity and inhibitory effects on food spoilage bacteria. Jinsixiaozao oligosaccharides were prepared efficiently by using 0.5 mol / L trifluoroaceticacid( TFA) to hydrolyze Jinsixiaozao polysaccharideat80 ℃ for 6 h,following by enzymatic hydrolysis of the resulting precipitation at 45 ℃ for 40 min by 0.05 mol / L pextaseon,and its yield was 24.87% ± 0.63%.The chromatographic conditions for analyzing of the monosaccharide composition were mobile phase,acetonitrile- water 28 ∶ 52( 0.1mol / L ammonium acetate buffer,p H4.3),flow velocity,0.8 m L / min,detection wavelength,254 nm. The results indicated that the Jinsixiaozao oligosaccharide extraction possessed inhibitory activities on bacteria,with minimal inhibitory concentrations on colibacillus,bacillus subtilis and S.aureusat 0.195,0.781,0.391 mg / m L respectively.The extract also had a certain antioxidant capacity,of which the total antioxidant capacity was( 4.69 ± 0.14) U,the DPPH·scavenging rate was 57.71% ± 0.51%,and the OH· scavenging rate was 35.24% ± 1.08% at a concentration of 25 mg / m L,respectively.All of these indicated that Jinsixiaozao had a value for development and utilization of its health- care functions.
